ObjectiveNeural regeneration and tissue's restoration is difficult when brain be injured。Drug treantment cannot have ideal effect。Permanent neural dysfunction maybe be usually remained。Recent research has found that transplanting MSCs to brain injured zone can also promote the restoration of brain's function。It indicated that transplantation of MSCs was a effective method to promote tissue recovery and improve neural function symptom。Nerve function depends on complex neural network.Ischemia can damage this structure.Studing the influence of MSCs to host's nerve fiber and the healing of neural dysfuction,which can help making it clearer the mechanism of which MSCs can promote the restoration of brain's function。MethodsSD rats were randomly divided into three groups,blank control group:modol of global forebrain ischemia rat;frozen MSCs-treated group:frozen MSCs were transplanted to double hippocampus' CA1 zone;MSCs-treated group:MSCs were transplanted to both hippocampus' CA1 zone.bone marrow stromal cells were isolated and cultured the in vitro,BrdU marked.Animal model of rat forebrain ischemia was made by occlusing four arteries.Blank control group were injected PBS;frozen MSCs-treated group were transplanted frozen MSCs to both hippocampus' CA1 zones;MSCs-treated group were transplanted MSCs to both hippocampus' CA1 zone.At corresponding time,Morris water maze test,immunohistochemistry,AchE and Timm's staining were performed.ResultsMSCs can quickly amplificate in vitro.The positive ratio of CD90 in immunocytochemistry is 98%or so.The positive ratio of BrdU marking is above 95%in vitro.After transplantation,MSCs can survive,a few of transplanted cells can express both BrdU and NeuN,both BrdU and GFAP.In the place navigation test,there is significant difference between MSCs-treated group and control group(p<0.01).There is difference between frozen MSCs-treated group and control group(p<0.05).At 12 weeks after transplantation,the AchE positive fiber density has significant difference in hippocampus' CA1 zone between MSCs-treated group and control group,between frozen MSCs-treated group and control group(p<0.01).Correlation analysis showed that the densities of AchE-rich fibers in hippocampus had significantly negative correlation with the escape latency in the place navigation test(r=-0.94).There is no difference among the three groups in Timm's stainingConclusionMSCs can quicklly amplificate in vitro.After transplantation,MSCs can survive,a few of transplanted cells can express both BrdU and NeuN,both BrdU and GFAP,which showed MSCs can transfer to neuron and glial cell.when MSCs be transplanted to brain injured zone,MSCs can protect cholinergic neurofiber.But it has no influence on moss fiber.Transplantation of bone MSCs to the forebrain ischemic areas can promote the improvement of the spatial learning.The mechanism which MSCs can promote the restoration of brain function maybe has the relation to the protection of cholinergic neurofibe and the microenviroment that benefit to function recovery.
