The Study Of The Active Compounds Of Zanthoxylum Nitidum And Application Of Oral Care Products

This study was performed to assess the active constituents of Zanthoxylumnitidum(Rox.b) DC on analgesia,anti-inflammation and hemostasia.We found neoherculin has significant efficacies on analgesia,anti-inflammation and hemostasia in mice.Nitidine chloride has some efficacies on anti-inflammation.The above research I have made will provide a scientific and reliable reference for the development and the application of Z.nitidum.The main content of this study as following:1.The biological activity of the extract fractions on analgesia,anti-inflammation and hemostasia were assessed:The extract of Z.nitidum(S-0) was separated into three fractions,S-1,S-2 and S-3. S-1 was the extract of S-0 by ethyl ether.S-2 was the extract of S-0 by 5%acetic acid,it is the total alkaloids.S-3 was the residuum of S-0 was extracted by ethyl ether and 5%acetic acid.The result of animal test showed that S-1 has significant efficacies on analgesia,anti-inflammation and hemostasia in mice,LD₅₀:68.85±10.31mg/kg.2.The constituents of the active extract fraction were isolated:By the chromatographed on a silica column,4 constituents were isolated from the active extract fraction(S-1),their structure were identified by ¹H NMR,¹³C NMR,¹H-¹H Cosy, HMQC,MS,IR and UV.They are①β-amyrin,â‘¡L-sesamin,â‘¢L-asarinin,â‘£noeherculin (0.28%).Noeherculin is riched in the Z.nitidum,it is pungent.By the way,⑤nitidine chloride(0.0088%) was isolated from the total alkaloids fraction.3.In Z.nitidum,we founded neoherculin has significant efficacies on analgesia, anti-inflammation and hemostasia in mice.Nitidine chloride has some efficacies on anti-inflammation in mice:The biological activity of the constituents of neoherculin and nitidine chloride were assessed on animal tests,the results show that neoherculin has significant effieacies on analgesia,anti-inflammation and hemostasia in mice,LD₅₀ 16.96±2.45mg/kg.Nitidine chloride has some efficacies on anti-inflammation in mice.This results will supply a scientific and reliable reference for the development and the application of Z.nitidum.4.The constituents of neoherculin and nitidine chloride etc.Of Z.nitidum growing in the ten areas of Guangxi were determined:Based on the noeherclin,nitidine chloride that were isolated from the active fraction (S-1) as the reference substances.We analysed and determined the content of the noeherclin,nitidine chloride in Z.nitidum at 10 different growing areas of Guangxi by high performance liquid chromatography(HPLC).The results show that the content of neoherculin was more different at the 10 different growing areas of Guangxi.The highest content of the neoherculin in Z.nitidum was 0.468%of Jinxiu;the lowest of it was 0.009%of Guilin.The content of nitidine chloride was also more different in the 10 different growing areas of Guangxi.The highest content of the nitidine chloride in Z.nitidum was 0.467% of Baise;the lowest of it was 0.049%of Jinxiu.This relults will supply a scientific reliable reference to purchase the better raw materials for the products of the toothpaste.And it is also a scientific reliable reference to choose the better growing areas for the plant of the Z.nitidum.5.The extraction of neoherculin from Z.nitidum:The effect of solvents,extract time,extract temperature on extracting neoherculin from Z.nitidum with reflux was studied with orthogonal design.The results showed that the content of neoherculin in the extract was varied from the extract conditions. When the roots of Z.nitidum was extracted by 75%alcohol,at 75℃for 3 hrs,the content of neoherclin in the extract was the highest(0.76%).When the roots of Z.nitidum was extracted by water,at 50℃for 1 hr,the content of neoherclin in the extract was only 0.00070%.6.The determination of the constituents of noehercline and nitidine chloride in toothpaste:According to the toothpaste prescription,we take the ratio of extract of Z.nitidum (S-0) to the materials of producing toothpaste,to make the products of toothpaste. Based on the noehercline and nitidine chloride as reference substances.We determinated them by the high performance liquid chromatography(HPLC).The results show that the content of neoherculin and the nitidine chloride can been determinated in the toothpaste.This relults will supply a scientific reliable reference data for the standard of this kind of Chinese herbal toothpaste.
