Tumors seriously threat to human's health and life, and the rate of tumor incidence and mortality increase year by year , Which becomes more and more obviously as ageing and industrial contamination. A report called the research of the prevalent tendency and prophylaxis about malignant tumor which was hosted by HaoXishan (the Chinese Academy of Engineering) discovers that the rate of fifty-nine kinds of malignant tumor was increased by 50% in the past 20 years from 1981 to 2000. Metastasis of the malignant tumor is one of the basic biological characteristics, which is the main reason of the majority of cancer patients leading to treatment failure and death. Therefore, in recent years, the importance of tumor metastasis has been paid more and more on academics. The course of tumor invasion and metastasis is a process of which primary tumor cells transfer to surrounding or distant tissues, involving tumor cells through extracellular matrix barriers, vascular basement membrane wall and entering into the host micro-environment .A large number of experiments show that the capacity of invasion and metastasis of tumor cells is closely related to protease degrading its extracellular matrix, basement membrane. Matrix metallo- proteinases can degrade almost all ECM components, which is the most important to tumor invasion and metastasis. In recognition of MMP in tumor invasion and metastasis plays such important role, therefore inhibitors of MMPs is also paid much attention to. Matrix metalloproteinase inhibitors (TIMPs) can inhibit not only the hydrolysis activity of MMPs, but also can inhibit the role of angiogenesis in tumor treatment, which is a promising research. At present, there is not enough evidence to prove whether the surgery, radiotherapy and chemotherapy treatment can control the tumor metastasis, while traditional Chinese medicine has unique advantages in improving quality of life of cancer patients, extending the survival time of patients with tumors and reducing the occurrence of distant metastasis.There are the following mechanisms in Traditional Chinese medicine : Firstly, six evils and exogenous evil including some chemical and physical factors invading the human body and influencing the run of Qi and blood leads to tumor. Secondly, anxiety, anger and grief lead to the stagnation of Qi, blood and sputum. Thirdly, the factors of the deficiency of Zang and Fu, the disorder of Qi and blood, exogenous evil, emotion , drink and food can easily hurt the human body. In a word, tumor has close relation to the weakness of the Zheng qi and the disorder of Zang and Fu, which is called"Together by evil, the gas will be virtual". Therefore, its treatment principle should be increasing the vital Qi to improve the capacity of anticancer and expel the evil Qi. The methods of warming the kidney yang, nourishing yin and blood can increase and consolidate the vital Qi. The other methods of Clearing away heat and toxic, promoting Qi and blood stasis can expelling the evil Qi. Traditional drugs have been a most important way of anticancer, and many research on them about the prevention and cure of tumor have been carried out. The characteristic of Hempseed is sweet and fatty ,the channel of it is spleen, stomach and large intestine ,the function is nourishing Zao ,lubricating the intestine and activating the blood . Modern research shows that hempseed has obvious therapeutic effect to the liver, stomach, cervical cancer, we can suppose that its treatment of tumors in vivo may be related to inhibition of matrix metalloproteinase hyper-expression. In this paper, hemp seed treatment study the mechanism of tumor will be studied.Objectives:To observe whether the extract of the hempseed can inhibit the matrix metalloproteinases , which offer essential theoretical and experimental foundation for the mechanism of the hempseed inhibiting tumor.Methods:Put a mount of MMP-13 solution to the enzyme buffer solution and the hempseed solution of 1ug/ml,10ug/ml,20ug/ml,40ug/ml differently , incubate 30 minutes ,then add 0.2μg DQ-gelatin and test the substrate .The fluorescence intensity can response the ability of enzyme degrading the substrate.Principles:The fluorescence can be emitted when the enzyme degrades the substrate , therefore testing the fluorescence intensity can response the ability of enzyme degrading the substrate. DQ-gelatin which is signed by the fluorophore is used in the experiment .After the enzyme degrades the substrate, the fluorophore is separated with quenching group .And to some extent, fluorescence intensity is proportional to the speed of enzyme degrading substrate. Results: The extract of the hempseed have the effect of inhibiting MMP-13 and the function depends on the density .That is to say , the hempseed extract on the inhibition of MMP-13 enhances With the increase in drug concentration ,which confirms the new drugs extracted from the natural and targeted to MMPs/TIMPs has bright prospects further.Conclusion:The extract of the hempseed can inhibit the activity of the MMP-13,and this kind of function depends on the density.
