The ovarian carcinoma becomes a serious threat to women's lives,and becomes a focus of research because of the difficulties in early detection. Vascular endothelial growth factor (VEGF) is a kind of protein that can specifically promote vascular endothelial cell growth and increases vascular permeability, which plays an important role in a variety of solid tumors in their occurrence and development.p53 gene is a tumor suppressor gene which has the highest mutation rate.p53 gene mutatation was detected in about fifty percents of human tumor tissues, wild-type p53 inhibites the expression of VEGF.These two factors affect the prognosis of ovarian carcinoma is don't work indepen- dently.They have intrinsic link with each other.In the vascular endothelial growth factor family,VEGF-C becomes a focus of research because of the close relationship with the tumor of female genital system .ObjectiveTo investigate the expression of vascular endothelial growth factor-C (VEGF-C) and p53 protein in ovarian epithelial carcinoma.To detect the relationship between p53 (VEGF-C) with clinicopathological factors and prognosis,the link with each other.To provide experimental evidence for early diagnosis and treatment of ovarian carcinoma.Materials and methodsImmunohistochemical method:Surgical specimens from 66 patients diagnosed by pathology in Obstetrics and Gynecology of the second hospital of Jilin university from February 2006 to August 2008 (including 32 cases with ovarian cancer, 10 cases with borderline ovarian tumor,14 cases with benign ovarian tumor,10 cases with normal ovary),collect ovarian tumor tissues in the surgery. All tissue samples were about the size of 1cm×1cm×1cm.To examine the expressin of VEGF-C protein,p53 by two-step immunohistochemical staining.Flow Cytometry:Fresh tissue samples from 48 patients diagnosed by pathology in Obstetrics and Gynecology of the second hospital of Jilin university from March 2008 to September 2008 (including 15 cases with ovarian cancer, 6 cases with borderline ovarian tumor,9 cases with benign ovarian tumor,10 cases with normal ovary), to cut and grind fresh tissue into single cell suspension.To detect immunofluorescence intensity of VEGF-C, p53 by flow cytometry.Results1.Immunohistochemical:(1)Expression of VEGF-C protein in epithelial ovarian tumors:Expression of VEGF-C protein in epithelial tissue of epithelial ovarian tumors:The middle-high level expression rates of VEGF-C protein is 71.86%(23/32) in ovarian cancers,20.00% (2/10) in borderline ovarian tumors. No middle-high level expression of VEGF-C is exhibited in benign ovarian tumors and normal ovarian tissues.Expression of VEGF-C protein in interstitium of epithelial ovarian tumors①Peripheral lymphocyte response to cancer and expression of VEGF-C protein, the relationship with metastasis of lymph node:Peripheral lymphocyte response to cancer relates to metastasis of lymph node(P<0.05),expression of VEGF-C protein in lymphocytes doesn't relate to it (P>0.05)。②Expression of VEGF-C protein in Vascular endothelial cells and intravascular nucleated cells (including monocytes and neutrophils).There is no significant difference in the expression of vascular endothelial cells of different groups. VEGF-C in intravascular nucleated cells of pithelial ovarian cancer and borderline ovarian tumors nucleated cells (including monocytes and neutrophils) show higher expression than in benign ovarian tumors,it shows significant difference(P<0.05). There is no significant difference in the expression of epithelial ovarian cancer and borderline ovarian tumors (P> 0.05)(2)Expression of p53 protein in epithelial ovarian tumors:The middle-high level expression rates of p53 protein is 59.38%(19/32) in ovarian cancers, 60.00%(6/10)in borderline ovarian tumors. No middle-high level expression of p53 is exhibited in benign ovarian tumors and normal ovarian tissues.(3)Relationship of expression of p53 and VEGF-C protein in ovarian cancer:There is positive correlation between VEGF-C and P53 protein expression(P<0.05).(4)Expression of p53 and VEGF-C in epithelial ovarian tumor and relationship with clinicopathological characteristics:The middle-high level expression rates of VEGF-C relates to clinical stage and lymph node metastasis. The middle-high level expression rates of p53 relates to histological grade, lymph node metastasis and ascites amount.2.Flow Cytometry :The immunofluorescence intensity of VEGF-C and p53 in ovarian cancer showed significant difference with benign ovarian tumor (P<0.05),there is significant difference in the rate of positive expression of p53 in single cell suspension of ovarian cancer,borderline tumor and benign tumor. The rate of positive expression in single cell suspension of benign tumor showed significant difference with borderline tumor and epithelial ovarian cancer ,There is no significant difference in borderline tumor and epithelian ovarian cancer.Conclusion1. Expression of VEGF-C protein can be used as an indicator to assess invasion of epithelial ovarian tumors.2. Peripheral lymphocyte response to cancer relates to metastasis of lymph node(P<0.05),it can be used as a factor to guide Pelvic lymph node dissection. VEGF-C protein expressed in lymphocytes ,but itdoesn't relate to metastasis of lymph node (P>0.05)。3. The Expression of VEGF-C in Nucleated cells (monocytes and PMN) in the blood vessels may be associated with tumor invasion and metastasis, and participate in formation of vascular, and antigen-presenting relevant.4. The expression of VEGF-C relates to clinical stage and metastasis of lymph node.No siginificant difference exhibits in different histological grade, the availability of planting pots and ascites amount.5. p53 in borderline ovarian tumors express higher than in epithelial ovarian cancer and benign ovarian tumors maybe because the accumulation of nuclear wild-p53 protein.6. The expression of p53 relates to histological grade, metastasis of lymph node and ascites amount. No siginificant difference exhibits in different clinical stage, and the availability of planting pots.7. There was positive correlation between VEGF-C and P53 protein expression.p53 can promote the secretion and expression of VEGF-C. |
