Study On Extraction And Quality Control Of Total Flavonoid In Euphorbia Humifusa

In this study,the method of extraction,abstraction,purification of total flavonoid in Euphorbia humifusa have been established,the method of determination of total flavonoid and quercetin also have been established.We estimated the content of total flavonoid and quercetin and studied on the finger print in different part of Euphorbia humifusa for its quality controlling.extracting,abstracting,purificating the total flavonoid in Euphorbia humifusa by the method of ultraphonic-heat recirculation-macroporous adsorptive resins,sieveing the best technology of ultraphonic extraction with the orthogonal test.The result indicated that 100g Euphorbia humifusa powder contains 3.131g pure flavonoid,it is 3.13%;The most optimal conditions of ultraphonic extraction is:the concentration of ethanol is 70%,the ratio of stock is 1:20,temperature is 60℃and extracting for 35min.four factors that impact the ultrasonic extracting is ultrasonic time＞solvent concentration＞stock ratio＞ultrasonic temperature.Established the method of ultraviolet spectrophotometry to Determined the total flavonoids (the rutin as the standard and the NaNO2-Al(NO3) 3-NaOH as the developer syetem, determine bsorbance at 510nm),the results indicated the standard curve of total flavonoids was linear in the range of 2μg-16μg(R=0.9933),the average recoveries is 102.2%.Established the method of HPLC to Determine the quercetin(chromatographic column: ShimADZUVP-ODS-C18,51μm,4.6×250 mm;temperature:35℃;detector:ultraviolet detector; wave:360nm;mobile:phase:methanol(50):0.4%phosphoric acid(50);flow rate:0.8 ml/min;inject: 10μl),the results indicated The standard curve of the quercetin was linear in the range of 0.5835μg-29.190μg(R=0.9998)and the average recoveries is 99.6%.for evaluating the contents of flavonoids in euphorbiae humifusae.the contents of total flavonoids and quercetin at different parts were Determined.the results indicated The contents of total flavonoids and quercetin at different parts of euphorbiae humifusae from high to low is:leaf, entire plans,seeds,caudex and roots,in leaf,The contents of total flavonoids and quercetin reached 43.25mg/g and 17.86 mg/g respectively,the study can be used to establish the standard of the quality for the total flavonoids and quercetin in different parts of euphorbiae humifusae.Studied on the finger print of different parts by HPLC,calculated similar degree of the HPLC chromatogram with the similarity evaluation software published by committee of codex, the Result display that the chemical composition are different between different parts,the similarity of leaf,roots,caudex and seeds with the entire gross is respectively 0.93,0.818,0.794, 0.751.
