Studying on compound hawthorn oral solution, a qualitation method of the composition and a quantitation method for the preparation have been established in this thesis; all these works supplied fundamental information for the optimization of extraction methods. The main work focused on the following aspects:1. Seven new extraction methods were designed compared with traditional method according to what were consulted in the database. The determination methods HPLC (high performance liquid chromatography) and UV spectrophotometry were applied to determine the quantity of stilbence glucoside and total flavonoids. The stilbence glucoside was detected with an Agilent Ecllpse, XDB-C18 column (5μm, 250×4.6 mm) by isocratic elution acetonitrile-water (22:78,V/V), and the total flavonoids with UV spectrophotometer at 510nm. The results showed that the two kinds of solution (NO.7,8) which Tuber Fleeceflower Root extracted separated contained most stilbence glucoside (108.6% and 102.7% were improved respectively), and when six herbs extracted together by diluted ethanol the solution (NO.3) contained most total flavonoids (66.9% was improved).2. The qualitation of the oral solution was made in chapter 3. The retention time of 38 components in the solution were determined according to the fingerprints, positive and negative ion signals were collected to assist the qualitation.3. The quantitation of the solutions which were extracted by five different methods was made in chapter 3, too. The samples were separated by an Waters Atalantis T-3 column, with gradient elution water (0.1% formic acid) and acetonitrile (0.1% formic acid). According to the signals by the negative ion detector, 28 components were determined and the relative quantitation was compared with each other. The solution (NO.7,8) contained most stilbence glucoside (130% and 110% were improved respectively), and NO.3 solution contained most total flavonoids.Methods that extracted Tuber Fleeceflower Root separated and six herbs extracted together by diluted ethanol were inferred as the excellent ones.
