| Objective To observe the expression of hepatocyte growth factor(HGF) in human umbilical vein endothelial cells(HUVECs) and the effect of ademosine on it to obtain a theory evidence that adenosine can provide protection effect when the cardia is experienced ischemia-reperfusion injury.Methods The cultured HUEVCs were divided into three groups according to the content of ademosine added to it: control group, experiment A group and experiment B group. The content of ademosine added to it were 0mmol/l, 10-4mmol/l and10-8mmol/l respectively. Immunohistochemistry staining was performed to detect the expression of HGF protein and RT-PCR was performed to detect HGF mRNA expression after 48hours.Results Immunohistochemistry study showed that there was only a small amount of HGF positive cells and the color was weak in control group.In experiment A group, HGF protein was significantly higher than that in control group(P<0.05).In experiment B group,there was no significant difference compared with control group(P>0.05).RT-PCR showed that the expression of HGF mRNA in experiment A group was higher than that in control group(P<0.05),while there was no significant difference between experiment B group and control group(P>0.05).Conclusion HGF is expressed in HUVEC and adenosine may promote HUVEC proliferation and angiogenesis partly through including HGF expression. |
