The Effects Of Adenosine On Expression Of Hepatocyte Growth Factor(HGF) In Human Umbilical Vein Endothelial Cells (HUVECs)

Objective To observe the expression of hepatocyte growth factor(HGF) in human umbilical vein endothelial cells(HUVECs) and the effect of ademosine on it to obtain a theory evidence that adenosine can provide protection effect when the cardia is experienced ischemia-reperfusion injury.Methods The cultured HUEVCs were divided into three groups according to the content of ademosine added to it: control group, experiment A group and experiment B group. The content of ademosine added to it were 0mmol/l, 10-4mmol/l and10-8mmol/l respectively. Immunohistochemistry staining was performed to detect the expression of HGF protein and RT-PCR was performed to detect HGF mRNA expression after 48hours.Results Immunohistochemistry study showed that there was only a small amount of HGF positive cells and the color was weak in control group.In experiment A group, HGF protein was significantly higher than that in control group(P<0.05).In experiment B group,there was no significant difference compared with control group(P>0.05).RT-PCR showed that the expression of HGF mRNA in experiment A group was higher than that in control group(P<0.05),while there was no significant difference between experiment B group and control group(P>0.05).Conclusion HGF is expressed in HUVEC and adenosine may promote HUVEC proliferation and angiogenesis partly through including HGF expression.
