Objectives:To find a method of measuring the Tissue Engineered Skin viability,and then use this method to comparing study on three kinds of preservation liquid,at last find a suitable preservation liquid to preserve the Tissue Engineered Skin under 4℃condition.Methods:First use the same kind preservation liquid to preserve two groups Tissue Engineered Skin,after preserved the same time,we use SYTO/EB method and MTT method to measure the skin's viability,and then analyse these datas with statistics to compare these two methods.Second,measure the viabilities of these skins preserved in 3 kinds preservation liquid(Ⅰ,Ⅱ,Ⅲ) with SYTO/EB method. And analyse these datas with statistics to find a suitable preservation liquid to preserve the Tissue Engineered Skin.Results:Through the study on SYTO/EB and MTT methods, we find there is no exact difference between these two methods on measuring the viability of the Tissue Engineered Skin.Under 4℃,after 1d,the viability of these skins preserved in those 3 kind of liquid shows no exact difference.But after 3days,5days,7days,10days and 14days,the viability of these skins preserved in liquidⅠand liquidⅡ,that liquidⅠand liquidⅢshows difference.After 14days,these skins preserved in liquidⅠkeep the best viability.Conclusion: The SYTO/EB double nucleic acid dye is a convenient,hypersensitive and rapid method of measuring the viability of skin,and the viability of any type of cells can be observed in situ.We establish SYTO/EB method of the Tissue Engineered Skin successfully.And the preservation liquidⅠresearched by our lab is the most suitable liquid to preserve the Tissue Engineered Skin under 4℃condition.
