Background and aimsIt's showed that the activation of hepatic stellate cell(HSC) was the key element in the progress of hepatic fibrosis. The proliferation of HSC induced by acetaldehyde was the key factor in alcoholic hepatic fibrosis. In recent years, many studies indicated that mitogen-actived protein kinase(MAPK) in culding ERK, JNK and p38 was one of the main signal pathway in the activation and proliferation of HSC. This experiment was to study the effects of mitogen-activated protein kinase antisense oligodeoxynucleotide(MAPK ASO)on the activation of acetaldehyde-induced hepatic stellate cell(HSC)biology characteristic.Methods After MAPK ASO was introduced into activated rat HSC by means of liposomal transfection, the proliferation of HSCs was evaluated by MTT assay, the cell colony formation test, the cell protein contain by Commssie Blue assay, the expression of MAPK-JNK pathway was examed by indirect immunoflurescence antibody technic and flow cytometry.Results: Compared with the control group,the mount of HSCs proliferation was inhibited by MAPK ASO,the inhibitory rate was 40.81%,the formation of cell colony was inhibited , the inhibitory rate was 39.9%,the cell protein contain was degraded,only 55.1%,the JNK signal pathway was significantly inhibited by MAPK ASO, the inhibitory rate was 60.6% respectively (p<0.05).Conclusions MAPK ASO exist important effect of certain biology characteristic on induced HSC, which maybe become an effective therapy for liver fibrosis. |
