Objective To observe Astragalus Membranaceus(AM) on effect of Peripheral blood mononuclear cells(PBMC) direvd-dendritic cells(DC) functions in children with Henoch-Schonlein Purpura(HSP),so offer theory base into its prevention or therapy with AM.Methods 28 HSP children in acute phase were detected,in which Peripheral blood mononuclear cells(PBMC) samples were obtained by density gradient centrifugation and divided into HSP group and AM group respectively,Then PBMC samples were induced with rhGM-CSF,rhlL-4 and rhTNF-αto make them transform into mature DC.The expression of the co-stimulatory molecules CD83,CD86(B7-2) and CD86(B7-1) on peripheral blood DC were examined by fluorescent activated cell sorter and the levels of IL-10,IL-12,IL-18 in supematant were measured by ELISA.Results①No significant distinction in the expression of CD83,CD86 and CD80 on peripheral blood DC was found between HSP group interfered with AM and HSP group(t或t'=0.37~1.49,P>0.05 ).②Compaied with HSP group,the supernatant levels of IL-12 in HSP group interfered with AM were higher[(141.58±100.19) pg/ml vs (96.18±76.65) pg/ml,t=3.90,P<0.01],while IL-10,IL-18 were both lower, respectively[(7.96+4.12) pg/ml vs(14.68±12.80) pg/ml,t=2.70,P<0.05]; [(15.78±5.66) pg/ml vs(37.94±29.47) pg/ml,t=4.07,P<0.01]。Conclusions The levels of IL-12 produced by DC increase after being interfered with AM,while IL-10 and IL-18 decrease.It is mainly through changing the levels of the cytokines secreted by DC that AM regulates its functions in children with HSP.
