Objective:To investigate the rule of expression of VEGF-C and VEGF-D and thire receptor Flt-4 in colorectal cancer,and to explore the relationship between them and the biological behaviour, the prognosis of colorectal cancer.Methods:The expression of VEGF-C/-D and Flt-4 in 45 cases with colorectal cancer was detected by immunohistochemical technique and its relationship with biological behaviour and prognosis of colorectal cancer were evaluated.Results:The expression of VEGF-C or VEGF-D was significantly higher in tumor tissue than that in normal tissue(P<0.05).The expression of Flt-4 was significantly higher in tumor tissue(44.4%) than that in normal tissue(6.7%P=0.008),there was a significantly positive relation between VEGF-C/-D expression and Flt-4 expression in tumor tissue(r=0.557,P<0.01;r=0.730,P<0.01).The expression of VEGF-C and VEGF-D and Flt-4 was significantly higher in the group with infiltration of lymphatic vessels than that without(P<0.05).The expression of VEGF-C and VEGF-D and Flt-4 also had significantly difference in the group with or without lymph node metastasis and different Dukes stage.With regard to VEGF-C and VEGF-D and FLT-4 expression,there was a positive relation to lymph node metastases(r=0.481,P<0.01;r=0.531, P<0.01;r=0.457,P<0.01),there was a nagetive relation to different Dukes stage(r=-0.552, P<0.01;r=-0.487,P<0.01;r=-0.410,P<0.01).In 45 cases,the postoperative five-year survival rate was 53.3%.The rate was lower in the group with negative expression of VEGF-C,VEGF-D and Flt-4 than that with positive expression of VEGF-C,VEGF-D and Flt-4(P<0.05).Conclusions:There was a closed relation between VEGF-C/-D and Flt-4 positive expression in colorectal cancer tissue,It indicated that every link of VEGF-C-Flt-4/VEGF-D-Flt-4 signal transduction path were all overexpressed.The overexpression may play a certain role in lymphatic vessels onset and promoting tumor cells metastasis of colorectal cancer;Through inducing tumor lymphatic vessels onset and promoting lymph node metastasis of colorectal cancer,the biological behaviour of colorectal cancer was effected by VEGF-C and VEGF-D.The detection of VEGF-C and VEGF-D expression may regard as a ancillary marker to predict lymph node metastasis of colorectal cancer;The high positive expression of VEGF-C and VEGF-D was correlated with the prognosis of colorectal cancer,and they may serve as one of molecule markers to evaluate prognosis of patients with colorectal cancer in the future. |
