Objective To determine the association between HLA classâ…¡genes and antithyroid drug(ATD)-induced leucocytopenia in patients with Graves disease.Methord The HLA-DRB1 alleles were typed by the polymerase chain reaction based sequence-specifec primer(PCR-SSP) method in group GDL(65 GD patients with ATD-induced leucocytopenia and 65 GD patients with leucocytopenia),group ATDL(65 GD patients with ATD-induced leucocytopenia),group NATDL,65 GD patients with leucocytopenia,NGDL,65 leucocytopenia patients without GD.group CON,(65 normol controls).And frequency of HLA-DRB1*08032 and DRB1*1501 gene expression were compared by x~2 test among groups.The risk of HLA-DRB1*08032 and HLA-DRB1*1501 gene expression were analyzed by binary logistic regression from all the patients' data.Results 1.In the allele frequency of HLA-DRB1*08032,there was no statistically significant difference between CON group and NGDL group(P=0.795),while it differed statistically significantly between CON group and GDL group(P=0.000),between CON group and ATDL group(P=0.000), between ATDL group and NATDL group(P=0.045).There were no statistically significant differences of sex differences between each groups in the allele frequency distribution of HLA-DRB1*08032.2.In the allele frequency of HLA-DRB1*1501,there was no statistically significant difference between CON group and NGDL group(P=0.795),while it differed statistically significantly between CON group and GDL group(P=0.000),between CON group and ATDL group(P=0.000).In the allele frequency of HLA-DRB1*1501,there was statistically significant difference between ATDL group and NATDL group(P=0.035).There were no statistically significant differences of sex differences between each groups in the allele frequency distribution of HLA-DRB1*1501.3.The factors that was significantly contributed to the ATD- induced agranulocytosis of GD patients is carrying the allele of HLA-DRB1*08032(P=0.034). Conclusion 1.GD with leucocytopenia were associated with HLA-DRB1*08032 and HLA-DRB1*1501 gene expression.ATD-induced leucocytopenia were associated with HLA-DRB1*08032 and HLA-DRB1*1501 gene higher expression 2.HLA-DRB1*08032 is the independent Risk factor of ATD-induced granulocytopenia and agranulocytosis.
