Experimental Study On Formula Fuzi Decoction By Semi-Bionic Extraction

Objective: To optimize the best Semi-Bionic Extraction(SBE) condition for Fuzi Decoction and futher verifying the correctness and practicality of Semi-Bionic Extraction(SBE).Method: Using the even designment to optimize the best extraction conditions of Semi-Bionic Extraction for Fuzi Decoction,with paeoniflorin,ginsenoside Rg1,ginsenoside Re,total aconitine,total saccharine,volatile oil,tolal integral calculus area of 50% alcohol solution by HPLC,dried extract weight as the indexs.To optimize the extraction methods of combined ingredients of Fuzi Decoction.According to the former conditions,the five kinds of extract metods,including the semi-bionic extraction(SBE),the water extraction(WE),the semi-bionic extraction by precipitation with alcohol(SBAE), the water extraction by precipitation with alcohol(WAE) and the alcohol extraction,were compared in the contents of the index ingredients and in the fingerprinting of the rabbit,s blood serum.Result: The technological conditions for SBE:the PH value of the water for the 1st,2nd,and 3rd decoctions are in order 6.0,7.5,8.5,and the extraction times are 2h,1h and 0.5h respectively.Mixing Aconitum carmichaeli Debx,Paeonia lactiflora Pall,Rhizoma Atractylodis Macrocephalae,Poria,Radix Ginseng together to extract is the best combinamic.The sequence of total index Y value of five kinds of alcohol precipitation concentration is: Y61.25%>Y55%>Y67.5%>Y80%>Y30%.Combined with the production,the best alcohol precipitation concentration is:60%.The sequence of total index Y value of four kinds of alcohol extraction concentration is: Y50%>Y30%>Y70%>Y90%.The sequence of total index Y value of the five technology is: YSBE液>YWE液>YAE液>YSBAE液>YWAE液.Using HPLC for completely comparing the ingredients of SBE and WE,the resuslt indicates that there has been no change in the nature of SBE and WE,but the content in SBE is obviouse high than in WE ,SBE is the best extraction method in preparation of Fuzi Decoction.Conclusion: Semi-Bionic Extraction method(SBE) maybe can replace Water Extract method(WE),Semi-Bionic Extraction method by precipitation with Alcohol (SBAE) maybe can replace Water Extract method and Alcohol precipitation(WAE). The practical condition of Fuzi Decoction with SBE has been optimized. This study lay the way for the future research.
