| Objective: 1. Exploring the optimum concentration and the mechanism of Acanthopanax senticosus injection inducing bone marrow stromal cells into neuron-like cell in vitro. 2. Studying the advantage of traditional Chinese medicine as inducer over Western medicine. Methods: 1. Inducing BMSC with different concentrations of Acanthopanax senticosus injection, measuring the proliferation of every groups of cells through MTT method and comparing the number of the positive cells by immunocytochemical method. 2. Inducing BMSC with the optimum concentration of Acanthopanax senticosus injection andβ-ME and comparing the number of the positive cells through morphological observation, immunocytochemical method and Nissl Body staining method. Results: 1. After induction, the differentiated cells showed to be typical neuron-like cells in all groups of Acanthopanax senticosus injection and (0.3-0.4)mg/ml was the best concentration which has little toxicity to BMSC. 2. The two groups of cells had no obvious difference after induction within 6 hours, but the number of the positive cells of the group of Acanthopanax senticosus injection increased with time extending and at the same time the number of dead cells is less thanβ-ME groups.Conclusions: (0.3-0.4)mg/ml is the optimum concentration of Acanthopanax senticosus injection which can induce bone marrow stromal cells into neuron-like cells in vitro and has less toxicity and better effect of long-time induction thanβ-ME groups. |
