| Objective: To investigate the relationship of a tumor related gene- DAB2 and tumor biological character in gastric carcinomas by detecting the protein and mRNA expression of DAB2, to more illuminate the pathogenesis of gastric carcinoma on the gene level, Aim to clarify the diagnostic and therapeutic value of DAB2 in human gastric carcinomas.Methods: The expressions of DAB2 Protein and DAB2mRNA were detected in the cancer tissues and its adjacent normal mucosa of 30 patients by immunohistochemical and RT-PCR method. Combined with clinical data, analysed the roles of the the expressions of DAB2 protein and mRNA in the development of the gastric carcinoma.Results: The DAB2 protein positive expression rate in gastric cancer and its adjacent normal mucosa is 26.7% and 86.7% respectively, the expression of the same tested index were found differed remarkably between two tissuss(p<0.05). DAB2mRNA expressed in both the cancer tissues and its adjacent normal mucosa, but its expression in cancer tissues(IOD ratio 1.100±0.284) is significantly lower than adjacent normal mucosa(IOD ratio 1.583±0.304) (p<0.05). Examination of the histories of 30 patients with primary gastric cancer revealed no statistically significant relationship between DAB2 protein and mRNA expression and the degree of tumor differentiation, the stage of the disease, lymph node metastasis , the age and the gender of the patients (p>0.05).Conclusion: As a tumor related gene, the positive expression of DAB2 in gastric carcinomas is significantly lower than that in normal mucosa, which testified DAB2 as a anti-oncogene and it may play an important role in the formation and development of gastric carcinomas. The expression of DAB2 have no correlated with the degree of tumor differentiation, the stage of the disease, lymph node metastasis in gastric carcinoma, the down regulatation of DAB2 expression may appear in early stage of the tumorigenesis, it has some clinical value in the early diagnosis of gastric carcinoma. There are not DAB2 protein expression in most gastric carcinoma, but all gastric carcinoma express DAB2 mRNA, it may have some posttranscriptional mechanism which effect the expression of the DAB2 protein, there are some value in illuminating the pathogenesis of gastric carcinoma by further studying its concrete mechanism. |
