Objective:To evaluate the association of the C242T polymorphism in p22phox gene of NAD(P)H oxidase with type 2 diabetes mellitus(T2DM) on Han and Korean population in Vanbian district of China,observe p22phox gene of NAD(P)H oxidase distribution characters in T2DM patients with atherosclerosis.Participants:Random select 681 cases in Yanbian district,including 120 Han and 102 Korea nationality people as the normal control group(normal glucose tolerant,NGT),273 Han and 186 Korean nationality patients as T2DM group.Patients were divided into four groups:①T2DM patients with carotid atherosclerosis of Han nationality(IMT≥1.0 and/or from atheromatous plaque);②T2DM patients without carotid atherosclerosis of Han nationality;③T2DM patients with carotid atherosclerosis of Korean nationality;④T2DM patients without carotid atherosclerosis of Korean nationality.Methods:Assess the NAD(P)H oxidase P22phox C242T polymorphism by PCR-RFLP(polymerase chain reaction -restriction fragment length polymorphism) analysis,diagnose carotid atherosclerosis by B-ultrasonic image.Statistical analysis with the 13.0 SPSS statistics software package,express data with mean±SE;analyze the genetype frequency with X~2 test;analyze the data with logistic regression to explore the independent risk factors of carotid atherosclerosis in T2DM patients(p<0.05 was considered significant) Results:(1) In normal control group,The NAD(P)H oxidase P22phox C242T gene distribution frequency in Han population is CC(77.5%),CT(20.4%),TT (2.5%),the allele frequency of C and T is 87.5%and 12.5%,in Korea population is CC(87.3%),CT(10.7%),TT(2.0%),the allele frequency of C and T is 92.6%and 7.4%(both P>0.05);(2) In T2DM group,The NAD(P)H oxidase P22phox C242T gene distribution frequency in Han population is CC (72.5%),CT(25.6%),TT(1.8%),the allele frequency of C and T is 85.3%and 14.7%,in Korea population is CC(84.9%),CT(12.4%),TT(2.7%),the allele frequency of C and T is 91.8%and 8.9%(both P<0.01):(3) There is no significant difference between normal control group and T2DM group in Han population(P>0.05),so as Korea population(P>0.05);(4) The gene distribution frequency of CC and CT+TT is 81.8%and 18.2%in T2DM patients with carotid atherosclerosis of Han nationality,67.2%and 32.8%in T2DM patients without carotid atherosclerosis of Han nationality;(5) The gene distribution frequency of CC and CT+TT is 89.2%and 10.8%in T2DM patients with carotid atherosclerosis of Korea nationality,82.1%and 82.1%in T2DM patients without carotid atherosclerosis of Korea nationality;(6) Logistic regression analysis shows that age is a risk factor which can induce carotid atherosclerosis in T2DM patients,B=0.914,OR=2.493,P=0.000 in Han population,where as B=1.005,OR=2.732,P=0.000 in Korea population. Conclusion:(1) There is a significant difference on p22phox gene of NAD(P)H oxidase distribution in T2DM patients between Han population and Korea population in Yanbian district.(2) T allele gene is a protective gene which can prevent carotid atherosclerosis,but it has no relation with T2DM;(3) The age is a strong and independent risk factor for carotid atherosclerosis in Han and Korean Population in Yanbian district of China.
