The mechanism of metastasis, however, is not completely defined. In the present study, we isolated two sublines of human osteosarcoma Saos-2 cells, which expressed higher E-cadherin protein than the primitive Saos-2 cells that presented only a weak expression. The E-cadherin expressed in these sublines made the cells more adhesive, and less motile and invasive. These observations demonstrated that among multiple regulation factors, E-cadherin, which promoted adhesion and inhibited migration in most carcinomas, could also affect the metastasis properties of osteodarcoma Saos-2 cells. We also study the effects of middle-low thermotherapy on expression and secretion of VEGF in SaoS-2 cells. Extract the whole protein and collect the supernatant of 43℃treated SaoS-2 cells seperatedly. And then the expression and secretion of VEGF in SaoS-2 cells were determined by Western blot and ELISA respectively. The expression and secretion of VEGF were both inhibited after 43℃treatment, and the inhibiton effects were enhanced as the treatment last.
