Objective:The purpose of this study is to observe the effect of celecoxib on inflammatory cytokine prostaglandin E2(PGE2) and interleukin-6(IL-6) secretion by cultured human gingival fibroblasts(HGF) in vitro and to investigate the effect of celecoxib on the periodontal inflammatory reaction.Methods:HGF were obtained from the gingival tissue adjacent to healthy premolars extracted for the orthodontic reasons.The tissue samples were placed into Dulbeccos Modified Eagles Medium(DMEM),cultured and subcultured at confluence.Experiments were carried out with cells from the fifth to eighth passages. HGF were palted in 96-well plates at an initial concentration of 2×105 cells/ml.After incubated for 24 hours,multiple concentrations of LPS 1,10,100,1000,10000ng/ml were added to the wells.The wells were incubayed for 6,12,24 hours.The supernatant were collected individually and then the concentration of prostaglandin E2(PGE2) and interleukin-6(IL-6) in the supernatant was assayed by ELISA to find the optimal stimulatory concentration of LPS. HGF were palted in 96-well plates at an initial concentration of 2×105 cells/ml.After incubated for 24 hours,multiple concentrations of celecoxib(0,50,100,200,400ug/ml) plus LPS(10000ng/ml) were added to the wells.The supernatant were collected after cultured for 12 hours. The concentrations of PGE2 and IL-6 were assayed by ELISA to obtain the optimal inhibitory concentration of celecoxib.Results:1,Under the stimulation of LPS significant greater amounts of PGE2 and IL-6 were released than those of the unstimulated cells.The production of PGE2 in HGF cells was increased in a time and does-dependent manner.The greatest amounts of PGE2 was at the 24 hours.But the greatest amounts of IL-6 was at the 12 hours.2,celecoxib can decrease LPS-Stimulated PGE2 and IL-6 production by human gingival fibroblasts.50ug/ml celecoxib can evidently decrease the production of PGE2 and IL-6 when 12 hours.conclusions:celecoxib can decrease LPS-Stimulated PGE2 and IL-6 production by human gingival fibroblasts, prevent the process of the periodontitis.
