SARS is a new infectious disease that the characteristic infected with sars coronavirus is present,and nowadays pathology of SARS hasn't been comprehended completely.Recovered SARS patients complications (avascular necrosis of femur head,pulmonary fibrosis)are frequent and serious problems.Recovered SARS patients complicating with avascular necrosis of femur head are complaint with the pain of local joint,even hip fracture;Recovered SARS patients complicating with pulmonary fibrosis are complaint with descent of exercise tolerances. These complications decreased terrible quality of patients'life.Nowadays,the reports on recovered SARS patients complications(avascular necrosis of femur head,pulmonary fibrosis)are often analysis on epidemiologic survey.The study on cause and pathogenesises of recovered SARS patients complications (avascular necrosis of femur head,pulmonary fibrosis) is just starting.Clinical diagnosis of recovered SARS patients complications (avascular necrosis of femur head,pulmonary fibrosis) always depend on complaint and imaging data of patients.It's too late to cure the diseases.So far, there is no effective way to early prevent and diagnosis of the diseases.With the rapid development of proteomic techniques in recent years,comparative proteomics has been obtained increasing applications and it offers us a new chance for screening specific biomarkers of diseases,studying the pathogenesis,and drugs'target.In this study, proteomic techniques were used to reveal the relative serum proteins of recovered SARS patients complications(avascular necrosis of femur head,pulmonary fibrosis) based on above ideas,explore the early diagnostic serum biomarkers,so as to provide scientific theory to effectively prevent,diagnosis and treatment of recovered SARS patients complications( avascular necrosis of femur head,pulmonary fibrosis).ObjectivesTo seek relative serum proteins of recovered SARS patients complications (avascular necrosis of femur head,pulmonary fibrosis),discuss the pathogenesis, so as to provide the theory to effectively prevent,diagnosis and treatment of recovered SARS patients complications(avascular necrosis of femur head,pulmonary fibrosis).Methods2-DE and MALDI-TOF MS were used to study on comparative serum proteomics among recovered SARS female patients complications (avascular necrosis of femur head,pulmonary fibrosis);ELISA detect serum amyloid P component and alpha-1-antitrypsin precursor in individual serum; Statistical software analysis specificity and sensitivity.Results 1.The 2-DE experimental maps of recovered SARS patients complications and health persons have a good repeatability and stability;2.Successfully discover 10 differentially expressed proteins including transthyretin,C4B3,fibrinogen gamma,apolipoprotein L,apolipoprotein A-IV precursor,albumin,prealbumin,serpin peptidase inhibitor,alpha-1-anti- trypsin precursor and serum amyloid P components.Contrasted to normal control group and simple group,transthyretin,C4B3,fibrinogen gamma, apolipoprotein L,albumin and prealbumin show lower expression (P<0.05);Contrasted to normal control group,simple group and fibrosis+necrosis group,serpin peptidase inhibitor ,alpha-1-antitrypsin precursor ,serum amyloid P components show higher expression in serum on necrosis group (P<0.05);3.The detected results of ELISA on the serum amyloid P component and alpha-1-antitrypsin precursor in the individual serum basically coincide with results of two-dimensional gel electrophoresis.Contrasted to normal control group,amyloid P component and alpha-1-antitrypsin precursor show higher expression in serum of necrosis group (P < 0.05) and fibrosis+necrosis group (P<0.05);4.The areas under the ROC curve on serum amyloid P component analysed by statistical software is 0.854, and the specificity is 77.8%,sensitivity of 85.2%;The areas under the ROC curve on alpha-1-antitrypsin precursor analysed by statistical software is 0.81,and the specificity is 75%,sensitivity of 69%.ConclusionRecovered SARS patients complications(avascular necrosis of femur head,pulmonary fibrosis)–relational protiens give a helpful hint to understand the molecular pathogenesis of recovered SARS patients complications(avascular necrosis of femur head,pulmonary fibrosis) deeply.Serum amyloid P components and alpha-1-antitrypsin precursor are possiblely potential biomarkers in serum of recovered SARS patients complications(avascular necrosis of femur head,pulmonary fibrosis). It is necessary to make a thorough study and discuss the results.
