A Preliminary Study On Expression Of CD4~+CD25~+Treg In The Peripheral Blood Of Hepatitis B Virus-infected Patients

Objective: To explore the expression of CD4~+ CD25~+Treg in the peripheral blood hepatitis B virus(HBV)-infected patients and its clinical significance,and to analyze the correlation between circulating CD4~+CD25~+Treg and serum HBV-DNA loads.Methods:1.Selection of asymptomatic carriers(ASC,n=35), acute hepatitis virus B(AHB, n=26), chronic hepatitis virus B(CHB, n=50) and health blood donators(n=30). Treg and T cell subsets(CD3~+,CD4~+,CD8~+) in peripheral blood of the four groups were detected by flow cytomerty .2.The hepatitis virus of antigen-antibodies were detected by ELISA in the patients with chronic hepatitis B and the normal controls.3.Hepatic function was detected by the automatic biochemistry analysator in all the objects.4.Real-time fluorescent quantitative PCR was used to measure the HBV DNA.5.Automatic coagulation analyzer was used to measure the serum prothrombin activity.Results: The expression of CD4~+ CD25~+Treg in the peripheral blood HBV-infected patients were : CHB group (12.71%±3.13%)>ASC group (11.32%±2.98%)> AHB group(8.87%±2.52%),the HBV-infected patients presented a higher fraction than healthy controls . CD4~+ CD25~+Treg expression in CHB and ASC elevated significally than that in healthy controls (P <0.01).However,there is little discrepancy between AHB and normal subjects. CD4~+ CD25~+Treg expression in CHB group and ASC group were positively correlation with HBV-DNA level(r=0.544,0.350, P<0.01),but not in AHB(r=0.35,P>0.05). CD4~+ CD25~+Treg expression in CHB group and ASC group were not positively correlation with serum ALT and AST level. CD4~+ CD25~+Treg expression in HBV-infected patients were not positively correlation with the level of CD3~+,CD4~+ and CD8~+( P>0.05) ,and there was positively correlation with the level of CD3~+ and CD4~+ in ASC group(r=-0.462,-0.470,P<0.01).Conclussion: The CD4~+ CD25~+Treg in ASC and CHB is increased.It could probably influence the of virus by supressing the antiviral T cell responses in CHB patients, Treg expression in CHB group and ASC group were positively correlation with HBV-DNA level .The finding suggested that CD4~+ CD25~+Treg play an active role in the HBV-infected patients ,and relate clinical development.
