| Objective:The gastric carcinoma is one of the most common malignant tumors in our country. The rate of incidence and death is the highest in all kinds of the alimentary tract malignant tumors. At present, the happening and development of gastric cancer is still emphasis of study. On the clinic, although about 60% patients with gastric carcinoma were through the valid treatment, they died of the recurrence and metastasis of tumor cells. The happening and development of gastric cancer is a progress in whichmany factors and processes and various mechanisms were involved.The aim of this study was to estimate the expression of FHIT and Ki-67 and to analyze their interaction in course of carcinogenesis of gastric cancer,to detect the pathogenesis ,progression and prognosis of gastric cancer.Methods:In this study, we select 50 samples of gastric carcinoma, 20 samples of normal gastric mucosa(>5.0cm). All patients had gastrectomy in the 4th hospital of HeBei Medical University from October 2006 to August 2007. All these samples are without chemotherapy or radiotherapy before surgery, which are proved adenocarcinoma by pathology. In this study, we use S-P immunohistochemical technique to detect the expressions of protein FHIT and Ki-67 in gastric carcinoma tissues, normal gastric mucosa and regional lymph node. The Correlations between the expressions of FHIT and Ki-67 and the biological behavior of gastric carcinoma were analyzed, including patients'sex, age, tumor location, size, gross type, differentiation degree, the depth of invasion, lymph node metastasis and TNM stage,clinical stage etc. All statistical analyses were carried out with SPSS12.0 and P <0.05 were considered significant.Results:1.The positive expression rate of FHIT gene was 95% and 34% in nomal gastric mucosa and gastric cancer. There was all significant difference beween the experimental groups(P<0.05).2. The positive expression rate of Ki-67 gene was 88% and 10% in gastric cancer and nomal gastric mucosa .There was all significant difference among the beween experimental groups (P<0.05).3. The positive expression rate of FHIT gene was 57.14% and 20.69% in tissues of high histologic grade of gastric cancer and low histologic grade of gastric cancer ,and was 70% and 22.50% in tissues of positive node metastasis gastric cancer and negative node metastasis gastric cancer ,and was 66.67% and 24.39% inâ… ï½žâ…¡stage andⅢ~Ⅳstage of TNM staging.There was all significant difference among the three experimental groups(P<0.05). 4. The positive expression rate of Ki-67 gene was 71.42 % and 96.55 % in tissues of high histologic grade of gastric cancer and low histologic grade of gastric cancer , and was 60 % and 95% in tissues of positive node metastasis gastric cancer and negative node metastasis gastric cancer,and was 66.67 % and 95.12% inâ… ï½žâ…¡stage andⅢ~Ⅳstage of TNM staging. There was all significant difference among the three experimental groups (P<0.05).5. No correlations were found between expression levels of FHIT and Ki-67 in gastric carcinoma tissues and patients'sexes, ages and tumor locations. (P>0.05).6. The reduction of FHIT gene expression might be positive related with the positive expression rate of Ki-67 gene in gastric cancer (r= -0.972, P<0.05).Conclusions:1.In gastric cancer tissues ,the low expression rate of FHIT gene and the high expression rate of Ki-67 gene may play important roles in the pathogenesis ,progression and prognosis of gastric cancer. FHIT and Ki-67 might become the molecular biological parameter to detect gastric cancer and its clinical biological behaviour.2. The expressions of FHIT in gastric carcinoma tissues become significantly lower along with differentiation degree to lower, the depth of invasion to deeper, lymph node metastasis, TNM stage to higher, however, the expressions of Ki-67 become significantly higher. That indicates that gastric carcinoma tissues become perniciouser,grow faster and obtin the higher ability of the invasion and metastasis.3. No correlations were found between expression levels of FHIT and Ki-67 in gastric carcinoma tissues and patients'sexes, ages and tumor locations. (P>0.05).4. In gastric carcinoma tissues, the expressions of FHIT and Ki-67 have negative correlation. It shows that the expressions of FHIT and Ki-67 in gastric carcinoma have the antagonism in invasion and metastasis of gastric carcinoma.In conclusion, the abnormal expressions of FHIT and Ki-67 in gastric carcinoma tissues play important roles in invade and metastasis of gastric carcinoma and the prognosis of gastric carcinoma. Examining the expressions of FHIT and Ki-67 in gastric carcinoma can be useful in prognosticating the invasion and metastasis of gastric carcinoma, evaluating the prognosis of gastric carcinoma and guiding the reasonable comprehensive treatment. |
