Section I The expression of WAVE1 in childhood acute lymphoblastic leukemia(ALL)Objective To investigate the roles of WAVE1 in the childhood Acute lymphoblasitic luekemia, we determined the expression of WAVE1 in myeloid tissue of normal children ,relapse ,complete remission and initial treatmental childhood acute lymphocyte leukemia.Methods Applying reverse transcriptase-polymerase chain reaction (RT-PCR) and Western bloting technique to semi-quantitatively analyze respectively WAVE1 and WAVE2 mRNA expressions in myeloid tissue from 10 normal children ,4 relapse ,15 complete remission and 40 initial treatmental childhood acute lymphocyte leukemia.Results Higher levels of WAVE1 expression were found in BMMCs derived from patients with primary and relapse leukemia. In contrast ,consistent with the absence of WAVE1 espression in BMMCs derived from normal healthy subjects ,or patients with complete remission.Conclusions Higher levels of WAVE1 expression were found in ALL. It is a novel monitoring target for pathogenetic condition of childhood ALL.Section II The expression of WAVE1 in the apoptosis of Jurkat cells induced by adriamycin(ADM)Objective To investigate the effects of ADM on the proliferation of human lymphoblastic leukemia (Jurkat cell line) in vitro.Methods 1,The changes of proliferation level of the Jurkat cells at 24,48 and 72 h after the treatment with ADM were detected by MTT assay.2,the rate of Jurkat cell with ADM apoptosis were observed by flow cytometry method in 24h;3,Applying reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting technique to semi-quantitatively analyze WAVE1 mRNA and protein expressions in Jurkat cell with ADM in 24h.Results1. 1,Following the prolonged time and different drug level, ADM could significantly inhibit the proliferation of Jurkat cells with a dose-dependent manner under the same duration of co-culture (after 24h 48 and 72 hours), p < 0.05;2,Following different drug level,the apoptosis rate is increased in 24hp<0.05;3,The espression of WAVE1 about mRNA and protein is dereased following the higher drug levels p<0.05.Conclusions1,When the ADM induce the apoptosis of the cells,the espression of WAVE1 about mRNA and protein is decreased.2,The WAVE1 in ALL may be play role in anti-apoptosis.
