| Objective This study is undertaken to approach the correlations between OPN and Caveolin-1 in pathogenesis and progression of osteoarthritis(OA)by detecting the expression of OPN and Caveolin-1 in different impaired cartilage of human OA.Methods To collect 36 cartilage specimens composed of OA(26) and normal joints(10).50 cartilage tissues with the form of 1.0 cm×1.0 cm×0.5cm were cut from these specimens according to the Outerbridge classification.All the cartilage tissues included normal and impaired cartilage ones.Then all the samples were ascribed to four groups of normal(A),minor(B),moderate(C)and severe(D)on the basis of the improved grading system of Mankin.The expression of OPN and Caveolin-1 were detected in these samples by the immunohistochemistry(SABC)and the expression result statistics were analysed by SPSS 13.0.Results 1.OPN were detected on the tissues of all the four groups.The amounts of average gray level of OPN expression were respectively(Group A)140.06±12.17,(Group B)125.33±8.02,(Group C)114.14±10.85,(Group D)98.32±12.91.The mutual comparisons of all these groups have statistical significance.(P<0.05).2.Caveolin-1 were also detected on the tissues of all the four groups.The amounts of average gray level of Caveolin-1 expression were respectively(Group A)85.90±15.11,(Group B)112.00±23.73,(Group C)131.72±19.32,(Group D)140.10±15.29.The mutual comparisons of all these groups have statistical significance(P<0.05) except the one between Group C and Group D(P>0.05).3.The average gray level of OPN in each group are accord with the improved grading system of Mankin(r=-0.824,P<0.01).That means the expression of OPN correlates with the grade of pathogenesis positively.The average gray level of Caveolin-1 in each group are also accord with the improved grading system of Mankin(r=0.725,P<0.01 ).It also means the expression of Caveolin-1 correlates with the grade of pathogenesis negatively.The negative corrlation between OPN and Caveolin-1 have statistical significance(r=-0.676,P<0.05).Conclusions 1.OPN play a role in the pathogenesis and progression of osteoarthritis(OA)2.The expression of Caveolin-1 were detected both in the normal and osteoarthric articular cartilage.The results show that the expression of comparison between normal and OA groups have statistical significance.The down-expression of OPN was obviously found with the company of the improving lesion of cartilage.It may indicate that Caveolin-1 inhabits pathogenesis and progression of osteoarthritis(OA)3.Caveolin-1 might defer the progression of articular cartilage through the down-regulation of OPN. |
