| Objectives: To study the expressions of vascular endothelial growth factor(VEGF) ,integrinαvβ3 and tissue factor in Brown Norway(BN) rats bydiode laser-induced choroidal neovascularization (CNV), investigating the effects of them in the progress of experimental CNV formation.Materials and Methods: Choroidal neovascularization was induced in BN rats bydiode laser. In every BN rat, one eye was the experiment eye which was induced CNV bydiode laser with the 532nm W/L. The power of laser was 500mW, the diameter of photo-coagulation spot was 50μm and exposure time was 0.05s. Fundus photography and fundus fluorescein angiography examinations were performed just before euthanasia on days 3,7, 14, 21 and 28 after laser photocoagnlation. The retina was processed for histopathology and immunohischemical analysis to detect VEGF, integrinαvβ3 and tissue factor.Results: There were disc like leakage of dioxyfluoran at the photo-coagulation area by fluorescein fundus angiography after 7 days later. VEGF was expressed in ganglion cell layer of retinal inner nuclear layer,retinal pigment epithelium and vascular endothelial cell of retina and choroid in normal BN rats. Integrinαvβ3 was scarely expressed in unactive vascular endothelial cell and other kinds of cells. VEGF,intergrinαvβ3 and tissue factor all expressed in ganglion cell layer of retinal inner nuclear layer,retinal pigment epithelium and vascular endothelial cell of retina and choroids in photo-coagulation area. The expression of integrinαvβ3 in rentina was decressed after 7 days(P<0.01) there are no significant differences of the expression of integrinαvβ3 between days 21 and 28. (P>0.05) With the development of CNV, the expression of VEGF and tissue factor expression were increasing (P<0.05), and be the strongest at 21day. The expression of VEGF and tissue factor had no significant differences between days 21 and 28.Conclusion: There were the expression of VEGF, integrinαvβ3 and tissue factor in the development of CNV. The strongest expressions of VEGF and integrinαvβ3 was at the earlier stage of CNV while tissue factor expressed at the later stage. The formation and development of CNV were related with the interaction of these three factors. |
