BackgroundParkinson's disease (PD) is one of the most common neurodegenerative disorders,the cause and pathogenesis of PD is complex, linking to aging, environment and genetic factors. The mechanism of dopaminergic (DA) neuronal degeneration and deletion remains unknown, oxidative stress, mitochondrial deficit and aggregation of misfolded proteins may play a role in it and any one of them could be the initial cause. Oxidative stress impairs mitochondria's function, causing proteins' misfolding and aggregation; some exogenous and endogenous toxins could tempt mitochondrial deficit, with a result in oxidative stress and protein misfolding and aggregation; Misfolded proteins are not simply poison to the DA neurons, but also induce oxidative stress and mitochondrial deficit.The interaction of them makes a vicious cycle and damage cascade, activating gitter cells, switching inflammatory reactions and causing neurotrophic factors insufficiency, which leading to the neuron's death. These intercrossing correlations raise a problem-- Are there some direct or indirect interactions between the PD associated proteins and whether they share a common way or network to play important roles in the differentiation, maturity, existence and function of the DA neurons. To date, bioresearch lays emphasis on single protein for its individual distinction and others which could interact with it. Now the problem is how to integrate these information based on monomers and investigate those interactions and their regulations with a more systematic and full-scale way. So, we use the complex network to get the integer character.ObjectiveTo construct the Network of Protein-Protein Interaction of Parkinson's Disease Associated Proteins; further to understand the function of these proteins and study pathogensis of PD; to introduce a simple but with affluent message way to analysis the Parkinson's Disease Associated Proteins.MethodsWe collect PD-related preteins and protein interactions by bioinformatics way, visualize and construct the Network of Protein-Protein Interaction of Parkinson's Disease Associated Proteins, calculate and analyze its detailed attribute.ResultsWe obtain a Network of Protein-Protein Interaction of Parkinson's Disease Associated Proteins containing 463 human proteins and 767 human proteins interactions;Index aggregation of the network is 90.5%,its p-value is 0.008, having statistical significance; we present a scoring method to rank proteins in the network,based on their contribution to the Network of Protein-Protein Interaction of Parkinson's Disease Associated Proteins and gain the relevance score of each protein of the network.Conclusions(1)We construct the Network of Protein-Protein Interaction of Parkinson's Disease Associated Proteins successfully by bioinformatics way;(2)discover some novel proteins may involving in PD pathogenesis;(3) introduce a simple but with affluent message way to analysis the bioinformation of Parkinson's Disease Associated Proteins.
