Objective To investigate the expression of matastasis associated gene 1 and membrane-cytoskeleton protein in breast cancer tissues and normal breast tissues.To study their correlation with the clinical stage, pathologic type,lymphnode matastasis and analyz their effect on matastasis and prognosis of breast cancer.To explore the possible moleculer mechanism of MTA-1.Methods 80 cases with breast cancer who were operated and tested by pathologic diagnosis in department of breast Xiangya Hospital of Central South University from March 2001 to March 2003 were selected for study groop,15 cases of normal breast tissue were selected for normal controls.The expressions of MTA-1mRNA and Ezrin in 80 cases of breast cancer and 15 cases of normal breast tissue were detected by in situ hybridization and immunihistochemistry SP method respectively.Analyzed their correlation with the parameters such as clinical stage,pathologic type,lymphnode matastasis et al,and studied their correlation with each other and effect on prognosis of breast cancer. Single factor was used by x2 test of crosstable and spearman's correlation to analyze prognostic fctors in primary breast cancer.Kaplan-Meier and Log-rank were used to test the influence of MTA-1mRNA and Ezrin on ratio of Disease-Free survival.Results1.The positive rates of MTA-1mRNA and Ezrin in normal breast tissues were 13.3%(2/15)and 20.0%(3/15)respectively,while those in breast cancers were 66.2%(53/80)and 68.7%(55/80)respectively.The expression of MTA-1mRNA and Ezrin were significantly different between breast cancers and normal breast tissues(P<0.05).2.The positive rate of MTA-1 mRNA in clinical stageⅢandⅣof breast cancers was 83.8%(26/31),while that in clinical stageⅠandⅡof breast cancers was 46.9%(23/49).The positive rate of MTA-1mRNA in the cases of lymphnode matastasis group was 86.9%(40/46),while that was 41.1%(14/34)in the non-lymphnode matastasis group.There were significant difference between the corresponding two groups(P<0.05). While the difference of the positive rates in different groups of age,tumor in size and pathological type were insignificant(P>0.05).3.The positive rate of Ezrin in clinical stageⅢandⅣof breast cancer was 87.0%(27/31),while that in clinical stageⅠandⅡof breast cancer was 48.9%(24/49).The positive rate of Ezrin in the cases of lymphnode matastasis group was 89.1%(41/46),while that was 47.0%(16/34)in the non-lymphnode matastasis group.There were significant difference between the corresponding two groups(P<0.05). While the difference of the positive rates in different group of age,tumor in size and pathological type were insignificant(P>0.05).4.The expression of MTA-1 in breast cancer was positiveiy related to Ezfin.(r=0.634,P=0.000).5.The ratio of Disease-Free survival in group of MTA-13+/Ezrin3+ was significantly lower than that in group ofMTA-1-/Ezrin-(x2=7.610, P=0.006)in breast cancer.Conclusion1.The positive rates of MTA-1 and Ezrin in breast cancers were significantly higher than those in normal breast tissues,which indicated there were correlations between the high expression of MTA-1 and Ezrin with occurrence of breast cancer. 2.The expressions of MTA-1 and Ezrin in breast cancers had not relations to ages,tumor in size and pathological types but to lymphnode matastasis and clinical stage.The higher expressions of MTA-1 and Ezrin indicated advanced clinical stage,higher tendency of matastasis and worse prospective.3.The united detection of MTA-1 and Ezrin expression in breast cancer could supply the evidence for judgement of prognosis.
