Objective With the coming of the aging of population in the world, it's an important research field about exploration of the mechanism of aging and how to delay the process of aging and exploring the effective prescription of preventing aging. Cynomorium songaricum Rupr(Cynomorium) is named as amrita drug. Its resistance is sweet and temperature, which has the functions of reinforcing kidney, strengthening yang, supplementing precision and so on. It has been used as an important aging drugs since ancient times. They are widely distributed in the Hexi Corridor and would become the developing potentiality Chinese medicinal materials in Gansu province. Our study was based on the basic theory and principles of Traditional Chinese Medicine. Through observing the effects of the chewable tablets of Cynomorium water extract and chewable tablets of Cynomorium polysaccharide extract on radical metabolism and immune function of the D-galactose (D-gal) subacute aging rats. We explored the anti-aging effects of chewable tablets of Cynomorium extract and its mechanism. The study contributed to the broad clinical application of Cynomorium chewable tablets and provides experimental evidence for delaying the aging process.Methods Wistar rats were randomly divided into normal control group, aging model group, vitamin E group,chewable tablets of Cynomorium polysaccharide group,chewable tablets of Cynomorium water extract group. All groups except normal control group hypodermic injection D-galactose seven weeks to established the aging model. Form second week, normal control group and aging model group were induced by successive perfusion of normal saline. Vitamin E group,chewable tablets of Cynomorium polysaccharide group,chewable tablets of Cynomorium water extract group which were induced by successive perfusion of vitamin E suspension, chewable tablets of Cynomorium polysaccharide group,chewable tablets of Cynomorium water extract group. After drug intervention six weeks, detected the activity of superoxide dismutase (SOD) by xanthine Oxidase technique,detected the content of malondialdehyde (MDA) by thiobarbituric acid test and the content nitric oxide (NO) w by nitrate reductase test in blood and brain tissue of rats in each group; Detected the proliferation activity of rat spleen lymphocytes stimulated by MTT assay and the spleen index, thymus index in each group. Observed the pathological morphological changes of brain and spleen;Results 1 .Compared with the normal control group, SOD activity of blood and brain of aging model rats were significantly lower,The content of MDA and NO of blood and brain of aging model rats increased significantly(P<0.05). The ageing rat model was made successfully. Compared with the model group, Chewable tablets of water extract and chewable tablets of Cynomorium polysaccharide improved significantly SOD activity of blood and brain of aging model rats; MDA content in blood and brain of aging rat model was reduced; Excessive NO increase of aging rats was decreased (P<0.05). Chewable tablets of Cynomorium polysaccharide group was superior than water extract group on improving the metabolism of of free radicals (P <0.05).2. Compared with the normal control group, the spleen index,thymus index, the proliferation activity of rat spleen lymphocytes stimulated by ConA of aging model rats were significantly lower (P <0.05). Chewable tablets of water extract and chewable tablets of Cynomorium polysaccharide could significantly increase the aging rat thymus index, spleen index and proliferation activity of rat spleen lymphocytes stimulated by ConA (P <0.05).3. The main retrograde degeneration of aging model group include: Neurocyte damages, hyperplasia of glial cell were appeared of the brain ,the white pulp and red pulp is not so distinct in spleen,the areas of white pulp reduced evidently, the red pulp introducing area and congestion. The groups of Cynomorium chewable tablets can improve the brain, spleen morphological pathological changes of aging rats.Conclusion Cynomorium chewable tablets has significant improvement on the free radical metabolism,immune function,pathomorphism changes of brain and spleen of D-galactose-induced animal aging model.
