| Objective To detect circulating tumor cells of breast cancer patients and to discuss its clinical significance by the MAGE-A1 and MAGE-A3 gene as specific marker. Methods Collecting cancer tissues ,surrounding tissues and peripheral blood of 60cases with breast cancer patients and peripheral blood of 60 cases with normal control group, The mRNA of the MAGE-A1 and MAGE-A3 gene was detected by nested RT-PCR.Results 28.3% (17/60) and 23.3% (14/60) of breast cancer patients PBMC were respectively detected to MAGE-A1 and MAGE-A3 mRNA. At least a MAGE gene mRNA was positive in the 35% (21/60) breast cancer patients PBMC. Detection rate of MAGE-A1 and MAGE-A3 mRNA in the PBMC of the breast cancer patients was related to the detection rate of MAGE in the tumor tissue. MAGE expression of the tumor tissue and peripheral blood was not related to tumor stage, tumor histological types, age, receptor status, CerbB-2, Ki-67 .Conclusion MAGE-A1 and MAGE-A3 may be used to detect the circulating breast cancer cells as the specific tumor markers, they could be specifically detected in the PBMC of breast cancer patients by our methods. |
