Objectives:To observe the effects of rosuvastatin and spironlactone on left ventricular(LV) remodeling and function in spontaneously hypertensive rats evaluated by echocardiography connected with the level of expression of myocardial collagen.Methods:Thirty-two male 16 weeks spontaneously hypertensive rats were randomly divided into four groups eight in each: rosuvastatin treatment group(group R) received rosuvastatin 10 mg·kg-1·d-1, spironolactone treatment group(group L) received spironolactone 20 mg·kg-1·d-1, rosuvastatin combined with spironolactone group(group R+L) received rosuvastatin 10 mg·kg-1·d-1 and spironolactone 20 mg·kg-1·d-1, and SHR control group. Tail systolic blood pressure of SHR were measured before treatment and two weeks, four weeks1, six weeks. left ventricular end-diastolic dimension(LVEDD) , intraventricular septal wall(IVS), left ventricular posterior wall thickness (LVPW) were evaluated by echocardiography before and six weeks after treatment. All rats were sacrificed after final evaluation for left ventricular mass and LV mass index(LVMI) measurements as well as histological study. Myocardial collagen were evaluated by using Van-Gieso staining and analyzed by image analysis system.Results:At time two weeks, four weeks and six weeks, blood pressure increased significantly in control group while decreased in group R, group L and group R+L and were lower than that of control group except for group L at time two weeks (P<0.05 or 0.01). LV mass and LVMI in group R, group L and group R+L were lower than their of control group. LEVDD decreased in control group while increased in group R, group L and group R+L after six weeks of treatment and were higher than that of control group(P<0.05 or 0.01). IVS and LVPW increased significantly in control group and decreased in group R, group L and group R+L except for LVPWS in group L(P<0.05 or 0.01). IVS and LVPW were lower in group R, group L and group R+L than their of control group(P<0.01). IVSD in group R were higher than that of group R+L and LEVDD in group L were lower in group L than that of group R+L(P<0.05). Myocardial collagen volume fraction (CVF) perivascular circumferential area(PVCA) in group R, group L and group R+L were lower than their of control group and group R+L were lower than their of group R, group L(P<0.01).Conclusions:1.Rosuvastatin and spironlactone therapy reversed left ventricular remodeling effectively in spontaneously hypertensive rats.2.Combined Rosuvastatin with spironlactone therapy have synergistic effects in reversed left ventricular remodeling in spontaneously hypertensive rats. |
