[Objective]To approach the capacity of erythrocyte regulating IL- 10 and IL- 18 in patients with acute cerebral infarction(ACI) by a experimental system of hemaimmune and the change of the natrue immunity of erythrocyte in vitro.[Methods]Patients with ACI 72h after onset were divided into two groups of the large,the midst and small by the volume of infarction and there was another group,which was the control group with dangerous factors but hadn't have a stroke.And there were 0.2ml suspension of cancer cells(S180:10×106/ml) or NS were added into 0.2ml anticoagulant suspension of whole blood cells or leukocytes and 0.3ml plasma,then incubated for 1 h at 37℃.The content of IL-10 and IL-18 were determined by ELISA.Then made groups comparison and the realationship of IL-10 and IL-18.[Results]1,In the patients with ACI,the IL-10 levels(pg/ml) in NEG(the large groupwas 5.68±2.60,the midst and small group was 3.58±0.84)were smaller than those in IEG.(the large groupwas 7.06±0.90,the midst and small group was 6.42±0.95)(P(0.01),but were larger than those in NCG(the large groupwas 2.27±0.86,the midst and small group was 2.43±0.84)(P<0.01).And the IL-10 levels(pg/ml) in the case groups were larger than those in ICG(P<0.01).The regulation rate of erythrocyte in the control group (-0.20±0.23) was smaller than those in the large groups(0.32±0.32) and the midst and small group(0.36±0.20) with cancer cells(P<0.01).And the IL-10 activated rates of erythrocyte in the large group(1.28±1.05) was larger than that in the control group (0.15±0.30)(P<0.01) while there were erythrocytes.2,The content of IL- 18 in the large group was much higher than the others(P<0.001).The regulation rate of erythrocyte in the large group(0.17±0.17) was larger than that in the control group(-0.16±0.32)(P<0.01) when there were cancer cells.While in whole blood,IL-18 activated rates of erythrocyte in the control group(0.63±0.61) was larger than in the large group(0.04±0.10)(P<0.01) and the midst and small group (0.21±0.31)(P<0.05).3,In the patients with ACI,the levels of IL-10 and IL-18 in NCG were negative correlation(r=-0.562,P<0.01).And the ration of IL- 18/IL- 10 was together with the serious of this disease.As to IL-10,there were some positive correlations between the groups,and so were that in IL-18 groups.[Conclusion]The results indicated that the experimental system of hemaimmune reaction,when used in detecting the regulative effect of erythrocytes to IL- 10 and IL- 18 secreted by leukocytes,was practical and convenient,in vitro.And it provid that the immunity of erythrocytes was decinded in ACI.And the regulative capacity of erythrocytes to IL-10 was postive,but to IL-18 was negative.And Then the regulative capacity of erythrocytes to IL-10 and IL-18 were concerned with the serious of the disease,and they were out of control while in the large group.The levels of IL- 18 was concerned with the square of the infarction.The levels of IL-18 and IL-10 was negative correlation in nature condition, which indicated that the imbalance between the two cytokines took part in ACI.And the ratio of IL-18/IL-10 was also concerned with he serious of the disease.It certified that the abnormalities and change of erythrocytes innate immunity represented a fundamental derangement in ACI.And at least there was one way that was by influenced the levels of IL-10 and IL-18.
