Chronic obstructive pulmonary disease(COPD)is characterized by incomplete reversible airflow limitation.The airflow limitation is gradually aggravated,which is concerned with the abnormal inflammatory reactions of lungs to harmful gas or granules.The incidence rate and fatality rate of COPD are high and COPD severely damage health.At present the treatment of COPD mainly through drug therapy,airway smooth muscle relaxation to help open lung airway and lower airway inflammation,mucus reduce the emergence of improved symptoms in patients with breathing difficulties and improve the quality of life.Bleomycin from Streptomyces round Health extracted a antineoplastic agents,it is one of the side effects caused pulmonary fibrosis in experimental pharmacology therefore can be used for pulmonary fibrosis model.Resveratrol is a polyphenol compounds in vivo and in vitro study showed that resveratrol has anti-inflammatory effect,but for the rare reports of COPD.In this paper,bleomycin smoked joint establishment of a new animal model of COPD,at the same time explore resveratrol on the intervention of COPD,and to investigate its possible mechanism.1.Establishment of chronic obstructive pulmonary disease models in rat by cigarette smoking and intratracheal instillation of bleomycinWe established COPD animal model by intra-tracheal instillation of bleomycin (BLM)once and exposure to cigarette smoke for continuous 27 days.At the 29th day,blood samples were taken from cervical artery for blood-gas analysis and parameters of lung function were recorded.Bronehoalveolar lavage fluid(BALF)was collected to measured intercellular adhesion molecule-1(ICAM-1)concentration.Our fingings show that the changes of pathology and lung function of this COPD rat model were well simulated the changes of COPD patients,decreased the ratio of forced expiratory volume in 0.3 second(FEV0.3)and forced vital capacity(FVC), and increased the ICAM-1 level in BALF.In addition,the COPD model in this experiment the new models of COPD was found. 2.Attenuation of chronic obstructive pulmonary disease in rats by resveratrolBecause resveratrol is insoluble in water,we use HP-β-CD as the hydrotropy at the rate of 1:10,and dissolve in normal saline.Everyday afer smoking,the rats inhaled atomized resveratrol(4,2ml·kg-1)20 min.At the 29thday,blood samples were taken from cervical artery for blood-gas analysis and parameters of lung function were recorded.Bronchoalveolar lavage fluid(BALF)was collected to measured intercellular adhesion molecule-1(ICAM-1)concentration.The results showed that atomization inhaled resveratrol could alleviated rat COPD lung injury accompanied by improvement of pathological changes,increased the ratio of forced expiratory volume in 0.3 second(FEV0.3)and forced vital capacity(FVC),and decreased the ICAM-1 level in BALF.The ultimate reduction of inflammatory factors was involved,at least in part,in the mechanism of resveratrol effects.In conclusion,our studies suggested that BLM and somoking compound method is better than classics model.It is reliable,controllable,short time.And resveratrol has the beneficial effect on pulmonary inflammation.Resveratrol deceased the the level of ICAM-1,had the function of protecting COPD.In addition,the irdaibtory effect of resveratrol on eytokines release,indicated resveratrol might have protective effect on lung inflammation by inhibiting excess expression of cytokines.
