Objective: To examine the variations in FCGR3B relative copy number of patients with lupus nephritis(LN),and investigate the relationship between FCGR3B relative copy number variations and clinical or pathological activity of LN.Method:202 patients with histologically proven LN and 146 geography-matched healthy control were enrolled from Peking Frist Hospital. Quantitative PCR was carried out using Taq man probes. The variations of FCGR3B relative copy number was examined with relative quantitative method. Clinical manifestations were collected among patients with LN, such as age, sex, urine protein excretion at the time of renopuncture, serum creatinine levels etc.The patients include classⅡ,classⅢ,classⅣand classⅤof pathological damage. SLE activity index was measured by SLE-DAI scoring to 190 patients whose clinical character was available.AI (activity index) and CI (chronicity index) were utilized on 123 patients whose pathological character was available to evaluate the pathological damage . Most important, to investigate the relationship between FCGR3B gene copy number variations and clinical or pathological severity of LN.Results: The distribution of FCGR3B relative copy number between group of LN and control was not being found with significant deffernce(P=0.627). Divided objects into groups by sex was no statistical difference(P=0.558,P=0.062). The distribution of FCGR3B relative copy number between group of classⅣ+classⅢand classⅡ+ classⅤwas also no statistical difference(P=0.511). No statistically significant difference in distribution of FCGR3B relative copy number was observed between SLE-DAI≤10 score and >10 score groups(P=0.996), AI<12 score and AI≥12 score groups( P=0.058), CI<4 score and CI≥4 score groups(P=0.401).Conclusion: In this study , the variations in FCGR3B relative copy number are not correlated with susceptibility of LN , and clinical or pathological activity of LN.
