Objective To detect the expression of melanoma antigen A1(MAGE A1)protein, melanoma antigen A3(MAGEA3) protein and NY-ESO-1 protein in human laryngeal squamous carcinoma and explore its significance in immunotherapatic application.Methods The expression of MAGEA1,MAGEA3 and NY-ESO-1 protein in the tissue of the tumor and in the pathologically positive lymph nodes were detected by PV-9000 Immunohistochemistry. The positive tissues were fluorescein stained and observed with Laser scanning confocal microscope. Western-blot was also employed to measure the expression of them in the tumor core region(TC) ,the region less than 0.5cm from the tumor edge(<0.5cm R), the region outside 0.5cm around tumor edge but less than 1.5cm (>0.5cm R), and in the distant normal larynx tissue.Results MAGEA1 protein expression was positive in 40 out of 69 (57.97%) cases of laryngeal squamous carcinoma, MAGEA3 positive in 20 out of 69 (28.99%) , NY-ESO-1 positive in 30 out of 69(43.5%), and 71.0% (49/69)cases expressed at least one of them, 39.1%(27/69 )cases expressed at least two of them, 18.8% (13/69)cases expressed all of them.The expression levels of three kinds of CTA protein were found to significantly decrease by turns in TC,<0.5cm R and >0.5cm R (P<0.01). None of the nine normal larynx tissues expressed them. It did not display an obvious correlation between the expression of the three kinds of CTA protein with T staging(P>0.05).The expression of MAGEA1 was significantly related to the pathological grading, and the expression of MAGEA3 in the cases with cervical lymph node metastasis was significantly higher than those without lymph node metastasis (P<0.05). Positive expression of three kinds of CTA protein was found in pathologically positive cervical lymph node, which was significantly lower than that in the primartumor (P<0.05). MAGEA1and MAGEA3 proteins were positive in cytoplasm under the LSCM, NY-ESO-1 positive in both nucleus and cytoplasm.Conclusions MAGEA1,MAGEA3 and NY-ESO-1 protien are expressed at high level in human laryngeal squamous carcinoma , and they may play a role in genesis and development of tumors, which suggests that these genes might be used as target antigens for immunotherapy of laryngeal squamous carcinoma.
