| Objective: To provide experimental foundation for clinical application by the observation of effect of jiaweixiaoyaosan which influences ethology and immune function in BALB/c mice being at chronic stress state.Methods: The stress model is made by putting BALB/c mice into a spicule pipe. The present experiment consisits of five groups:A(the normal comparison group), B(the model group),C(the xiaoyaosan group),D( the meta-dose jiaweixiaoyaosan group),E( the homo-dose jiaweixiaoyaosan group). The ethology indexes in mice are measured by the open-field experiment and hunging-tail exiperiment. To observe the changes of immune function of mice, the thymo-index and splen-index are measured , the content of CD4+ and CD8+of T cell are determined by using flow cytometry , IgG, IL-4 and IFN-r in blood serum by ELISA means.Results: Statistical analysis indicated that : 1.The ethology indexes:In the open-field experiment , the frequency of threading square of mice in group C,D,E is more than in group B(P<0.05)and the time when mice stays in the central square in group C,D,E is much shorter than in group B(P<0.05). 2. The immunology indexes :the spleen-index in group C,D,E visibly raises more than in group B(P<0.05), but the thymo-index in group C,D,E is similar to each other at statistic(P>0.05). The number of CD4+ cell in group C,D,E remarkably increases more than in group B(P<0.05) and the number of CD8+ cell in group C,D,E remarkably decreases more than in group B(P<0.05), and accordingly the ratio of CD4+ and CD8+ in group C,D,E visibly raises more than in group B(P<0.05).The value of IgG in group C,D,E obviously rises more than in group B(P<0.05)and IgG in group E is obvious. The value of IFN-r in B group is lower than group A,C,D,E(P<0.05),There appears no significant difference among group A,C,D in the value of IFN-r (P>0.05),the value of IFN-r in group E is more than group C(P<0.05);The value of IL-4 in group B is more than group A,C,D,E(P<0.05);There appears significant difference between group A and group C,D(P<0.05),and there is also no significant difference between group A,E in the value of IL-4(P>0.05).Conclusion: Chronic stress could cause abnornal of ethology and immunological regulating function. Xiaoyaosan or jiaweixiaoyaosan could remarkably rivalry the changes of ethology and immunological regulating function(The homo-dose jiaweixiaoyaosan group is better ). |
