Headspace Solid Phase Microextraction (HS-SPME), Solid Phase Extraction (SPE) and traditional Liquid to Liquid (L-L) Extraction gas chromatograph-mass spectrometry (HS-SPME-GC/MS) methods for qualitative and quantitative determination of several kinds of drug abuse such as 2C-B, nimetazepam, nitrazepam, midazolam, tramadol, methadone and amphetamines in biological samples were developed. Wistar rats and human being were selected as biological modes, administrered drug abuse and 24 h urine or tissues were collected. Tramadol and methadone were extracted using HS-SPME, the various parameters including extracting time, extracting temperature pH value and ionic strength were investigated for optimization of HS-SPME performance. 2C-B, nimetazepam, nitrazepam, midazalom and their metabolites were extracted using SPE from samples. Tramadaol, amphetamines and their metabolites were extracted using L-L rapidly. The several kinds of drug abuse mentioned above and their metabolites were qualitative or quantitative using GC/MS. The findings suggested the metabolic pathway of 2C-B, nimetazepam and nitrazepam and the corresponding mass spectrum data were created. The methods of quantitative determination of tramadol, methadone, amphetamines were developed. The proposed methods have been successfully applied to practical daily work.
