Objective Quantification detect the expression level of CK20 mRNA in the peripheral blood by fqRT- PCR around postoperative adjuvant chemotherapy in patients with gastric cancer,To investigate whether CK20 mRNA expression level could be considered as an effective molecular method for evaluation of chemotherapy sensitivity,this study was designed to explore the feasibility of this issue,and then could be provide important evidence for individualized chemotherapy.Methods All samples of peripheral blood were taken from each gastric cancer patients that had undergone an radical operation at the day before postoperative chemotherapy a week,chemotherapy point,after the first cycle of chemotherapy and subjected to fqRT-PCR assay for CK20 mRNA.The chemotherapy scheme was FOLFOX 4. The control group was 15 cases healthy volunteers.Results(1)The percentage of the CK20 mRNA expression rate increased from 61.3%before chemotherapy to 74.2%in chemotherapy point,and decreased to 58.5%after the first cycle of chemotherapy,there was no statistically significance between CK20 mRNA expression rate in these three groups(P= 0.373).(2)the value of CK20 mRNA before postoperative chemotherapy was increase(1. 76 vs 2.30,P=0.047),and then decline after chemotherapy(2.30 vs 1.59,P=0.033). (3)The expression level of CK20 mRNA in patients before chemotherapy was increased in 16 cases,declined in 9 cases and stabilized as negative in 6 cases.Afer chemotherapy the level of CK20 mRNA was increased in 7 cases,declined in 17 cases and stabilized as negative in 7 cases,there was statistically significance between change in these two groups(χ~2=6.06, P<0.05),and the level was increased in most of cases before chemotherapy(16/31),but after chemotherapy declined(17/31),χ~2=5.97,P=0.015.Conclusions The expression level of CK20 mRNA in the peripheral blood detected by fqRT- PCR in patients with gastric cancer declined after postoperative adjuvant chemotherapy,dynamic monitoring CK20 mRNA should be considered as an effective index to evaluate the efficacy of postoperative adjuvant chemotherapy.
