Objective To investigate the serumal neuroglobin fluctuation during 24h in ratssevere pancreatitis model, the pathological changes and distribution of Ngb in brain, and further estimate the correlation between neuroglobin fluctuation and brain damage in severe acute pancreatitis. Providing a new serumal diadynamic criterion, to lend support to evaluation of severity and diagnosis of pancreatic encephalopathy(PE).Method Experimental SD rats were divided into 3 groups: SAP (n = 24); MAP (n = 24) and Sham group (n = 16). Sodium taurocholate (STC) was injected under capsule of pancreas to make the model of SAP and MAP. Sham group was treated as protocal. Serumal Ngb level at 4h, 8h, 16h and 24h after model incubation was assayed using ELISA;Immunohistochemical methods were used to investigate the distribution of Ngb positive cells in rats brain of each group. Quantitative image analysis software was used to perform statistical analysis.Results SAP and MAP models were duplicated successfully. Serumal Ngb level was elevated remarkably at 4h after the SAP model incubation (P<0.05); It increased continuously during the following incubative time and significantly increased at 24h(P<0.01). Ngb positive cells increased with the aggravation of brain damage.Conclusion In early stage of SAP, Ngb which increases in brain may perform the neuroprotective effect in hypoxia condition, and the significant increase of serumal Ngb level indicates that to measure Ngb in serum is a meaningful method to make early diagnosis of PE.
