This dissertation included three parts:summary of related literatures; study on the chemical constituents of Woodwardia japonica from Guangxi;the quantitative determination of Ponasterone A and Ponasteroside A by High performance liquid chromatography(HPLC).Part one:The documents about the present research situation in Chinese medicine Gou Ji Guan-Zhong were summed up and analyzed.A review on the research progress of genus Woodwardia in chemical constituents and pharmacological activities was presented.Part two:Studies on the chemical constituents of Woodwardia japonica.Several separation and purification methods and determination technology were used,the research about chemical constituents of Woodwardia japonica was done,9 compounds were isolated and identified which include 1 triterpene,it is Woodwardic acid;2 flavonoids,they are kacmpferol-3-O-α-L-rhamnopyranoside-7-O-α-L-rhamnopyranoside and kaempferol-3-O-α-L-(4-O-acetyl)-rhamnopyranoside-7-O-α-L-rhamnopyranoside; 4 sterones,they are Ecdysterone,Achyranthesterone A,Ponasterone A and Ponasteroside A and 2 others, they areβ-sitosterol-3-O-α-L-(6-O-hexadecanoyl)glycoside and Daucosterol. Among them there were two constituents firstly separated from Woodwardia plants: Achyranthesterone A and Ponasteroside A;and two constituents firstly separated form Woodwardia japonica:Ecdysterone and Ponasterone A.And firstly a mixture was identified as kaempferol-3-O-α-L-(2,3-O-diacetyl)-rhamnopyranoside-7-O-α-L-rhamnopyranoside and kaempferol-3-O-α-L-(2,4 -O- diacetyl)-rhamnopyranoside - 7 -O -α- L- rhamnopyranoside.Part three:Establishment of the quantitative determination of Ponasterone A and Ponasteroside A by HPLC.The quantitative methods of indicative constituents Ponasterone A and Ponasteroside A by HPLC were established for the first time.The linear regression was respectively:y=688825x+11790(r=0.9995), y=542429x+2468.2(r=0.9999).The average recoveries of Ponasterone A were96.05%, RSD=0.62%;the average recoveries of Ponasteroside A were 95.73%,RSD=0.64%.The determination results of the contents of the two constituents in the plants from six different provinces showed that the methods were steady and reliable and can be used as the determination methods.
