| Background and ObjectivesCoronary heart disease(CHD)is a kind of multigenic diaease.With the rapid development of human genome project,the orientation and identification of CHD-related genes have been hot issues of medical research.Pathophysiologic research demonstrates that the formation of atherosclerosis correlates with endothelial dysfunction,and endothelial NO synthase(eNOS)plays an important role in the regulation of endothelial function.As a target kinase of small GTP-binding protein RhoA,ROCK2 can inhibit the formation and activation of eNOS.Furthermore,it can also upregulate the mediators of inflammation,enhance the vasoconstriction,promote the migration and proliferation of vascular smooth mucsle cells(VSMC).All these result in the evolvement of atherosclerosis.Resarchers have discovered ROCK2-125592 A/C polymorphism is associated with human hypertension disease, while less work has been done on its relationship with CHD.This study purposes to investigate the distribution of ROCK2-125592 A/C polymorphism in Chinese population and study its correlations with CHD.Materials and methods1.Subjects CHD group:263 CHD patients(191 male and 72 female,mean age 65±10.11 years)were consecutively(from March 2006 to December 2006)collected from the hospitalized patients in the department of cardiology,Sir Run Run Shaw Hospital affiliated to medical college of Zhejiang University.All patients were confirmed CHD by coronary angiography.Control group:211 CHD(-)objects(150 male and 61 female,mean age 64±10.54 years)served as a control group.All subjects were consecutively(from March 2006 to December 2006)collected from the hospitalized patients in the department of cardiology,Sir Run Run Shaw Hospital affiliated to medical college of Zhejiang University.All patients were confirmed CHD(-)by coronary angiography.2.Laboratory methods for detecting ROCK2-125592 A/C polymorphism1)DNA was extracted from the peripheral blood leukocytes by standard phenol/chloroform method.2)ROCK2-125592 A/C polymorphism was detected by polymerase chain reaction and gel-based DNA microarray method.3.Statistic analysisThe SPSS15 statistic software package was used.Difference between measurement data were performed by chi-square test.Genotype and allele frequencies between groups were counted and analyzed by chi-square test.A p value of less than.05 is considered to indicate a significant difference.Results1.We detected three kinds of genotypes at the ROCK2-125592 locus,and no deviation was observed from Hardy-Weinberg equilibrium.(p>0.05)2.The ROCK2-125592 A allele was detected in 34.1%of normal controls and 52.8%of CHD patients,while ROCK2-125592 C allele was detected in 65.9%of normal controls and 47.2%of CHD patients.There was significant difference in ROCK2-125592 allele and genotype distribution between CHD patients and normal controls(p<0.05).3.The genotype distribution of ROCK2-125592 A/C showed no difference between single,double and triple vessel lesion in CHD patients(p>0.05).4.Occurrence of risk factors,including Diabetes Mellitus,obesity, hyperlipmia,didn't show significant difference between 3 kinds of genotypes. (p>0.05).Conclusion1.The ROCK2-125592 A/C polymorphism exists in the Han population of Zhejiang province,China.2.ROCK2-125592 A allele may be a predisposing gene of CHD.3.ROCK2-125592 A/C polymorphism may not be associated with the degree of CHD.4.ROCK2-125592 A/C polymorphism may not be associated with Hypertension, Diabetes Mellitus,Obesity,or Hyperlipmia. |
