| After ischemic-reperfusion, the nerve cells died in form of necrosis or apoptosis. The necrosis of nerve cells in the center of ischemic area is irreversible; but the apoptosis of nerve cells in ischemic penumbra is reversible. So ischemic penumbra is a hot spot studied in ischemic cerebral vasocular diseases. Although the tissues of ischemic penumbra lost their physiologic functions temporarily, the cellular integrity was not injured and the cells were still alive. This research was designed to study these changes and the role of Piper kadsura Ohwi in focal cerebral ischemia .Objective:1. To observe nerve cell apoptosis, changes of DNA damage, SOD activity,MDA concentration and MMP after ischemia-reperfusion in rats.2. To investigate the effect of the extraction from Piper kadsura Ohwi on never cell after focal cerebral ischemia in rats.Methods:1. The rats of middle cerebral artery occlusion and reperfusion were used as the research model.2. To investigate DNA damage after focal cerebral ischemia-reperfusionand intervention of extraction from Piper kadsura Ohwi by single cell gel electrophoresis .3. To investigate the nerve cell apoptosis and MMP of nerve cell afterfocal cerebral ischemia-reperfusion for 2 hours through flow cytometry .4. To investigate SOD activity and MDA concentration 6, 24 and 72 hours after cerebral ischemia-reperfusion.5. According to the tranditional manufacturing methods of Chinese drugs, extraction of Piper kadsura Ohwi was made up and injected into rats abdominalcavity. Then, we observed its influence on those indexes mentioned above.Results:1. After the damaged DNA untwisting, small DNA fragments were released, and moved towards positive pole through the gel, coming to appear comet formation.2. Compared with sham operated group, the SOD activity decreased obviously 24hours and 72hours after ischemia-reperfusion, but with MDA concentration increased.3. Compared with sham operated group, the apoptosis increased obviously, accompanying MMP decreased.4. After intervention of Piper kadsura Ohwi, the apoptosis of nerve cell, DNA damage and the production of free radical all decreased obviously at different time in ischemic- penumbra .At the same time it also could stable MMP.Conclusion:Extraction from Piper kadsura Ohwi plays a significant role in protecting cerebral tissue through restraining oxidative stress after cerebral ischemia, decreasing the apoptosis and the production of free radical, and through stoping decrease of MMP of nerve cell caused by ischemia-reperfusion. |
