| Angelica sinensis and Astragalus have been widely used in traditional Chinese medicine. The determination methods and extraction process of polysaccharides from Angelica sinensis and Astragalus was investigated. The results are as following.Phenol-sulfuric acid and 3-hydroxydiphenyl method method were evaluated through the indexes as linearity and range, precision, stability, and accuracy and so on. Thus, it offered a significant reference in establishing the quality standard of polysaccharides from Angelica sinensis and Astragalus. The contents was determined with phenol-sulfuric acid method, and the contents of polysaccharides from Angelica sinensis and Astragalus were 11.31%,8.27% respectively.The optimum extraction process to extract polysaccharides of Angelica sinensis was determined by single factor test and orthogonal experiments.The optimum parameters to extract were as follows:ratio of water to material 10:1, four times of reflux extraction at 100℃, each for 2h. The best precipitation condition was as follows:at the concentration of 0.75g herb/ml water, three times of 95%alcoho1,12 hours of precipitation. The optimum condition of TCA method was studied, and the result was as follows: 5%TCA, removing 60 minutes for three times. The best condition of ultrafiltion was gained through experiment: 0.09 MPa at room temperature. The contents of neutral sugar, uronic acid and total carbohydrate in polysaccharides of Angelica sinensis extracted and purified by this process are 25.16%, 41.64%, and 66.8% respectively, and the polysaccharide yield is 5.51%.The same methods were used to extract and purify Astragalus polysaccharides, and the optimum parameters were as follows:ratio of water to material 10:1, four times of reflux extraction at 100℃, each for 2h; at the concentration of 1.5g herb/ml water, three times of 95%alcohol,16 hours of precipitation; 5%TCA, removing 60 minutes for three times; the best pressure is 0.09 MPa at room temperature. The content of total carbohydrate in Astragalus polysaccharides extracted and purified by this process is 69.87%, and the polysaccharide yield is 1.41%.Based on the reliable research methods, extraction process that suit for Angelica sinensis and Astragalus were gained, and the work provided foundation for their further research and industrialized production. |
