| Objective: To investigate the therapeutic effects and the possible mechanisms of Pongamia pinnata root flavonoids (PRF) on the experimental ulcerative colitis.Methods:(1) The rat model of ulcerative colitis was established by clysis with acetic acid solution, the sulfasalazine(SASP)group was considered as positive control, the general symptoms and the lesion of colonic mucosa of the rats were observed; (2) The mouse model of ulcerative colitis was established by administrating ad libitum 4% dextran sulfate sodium(DSS)solution in distilled water for 7 days , the SASP group was considered as positive control, the general symptoms and the lesion of colonic mucosa of the mice were observed;(3) Antidiarrheal effects of PRF were evaluated through the experiments of castor oil and senna's effect on the small intestine , normal small intestine propulsion , antagonize neostigmine and all gastrointestinal tract propulsion;(4) The activity or content of MPO,SOD,NO and iNOS in the colonic tissue of rats with ulcerative colitis induced by acetic acid were examined to explore the mechanisms of therapeutic effects by biochemical examination;(5) The level of T-AOC and MDA in the colonic tissue of mice with ulcerative colitis induced by DSS were examined to explore the mechanisms of therapeutic effects by biochemical examination;(6) The mice models of ulcerative colitis were established by administrating ad libitum 4% DSS solution in distilled water for 7 days, the apoptosis of colonic epithdial cells were detected by TUNEL and the expression of Fas/Fasl were detected by immunohistochemistry (SABC) .Results: (1) PRF could remarkably improve the general symptoms and relieve the lesion of colonic mucosa of UC animal models; (2) PRF could obviously reduce the frequency of purging induced by castor or senna in mice, markedly inhibit the normal small intestine propulsion,reduce the influence of small intestine propulsion after hypodermic injection of neostigmine and prolong stools-excretion time in mice;(3) PRF could notably decrease the activity or content of MPO,NO,iNOS and increase SOD activity in the rat colonic tissue with ulcerative colitis induced by acetic acid;(4)PRF obviously decrease the content or activity of MDA and increase the level of T-AOC of colonic tissue of the mice with ulcerative colitis induced by DSS ; (5) PRF significantly reduce the apoptosis of colonic epithdial cells and the expression of Fas/Fasl in the colonic tissue of mice with UC.Conclusion: PRF has significantly therapeutic effect on ulcerative colitis and antidiarrheal effects. It may be a potent candidate in prevention and therapeutic drug of ulcerative colitis , and the mechanisms of anti-ulceractive colitis are possibly related to the following factors:①Inhibiting inflammatory cell infiltration of colonic tissue;②Modulating the oxidant-antioxidant imbalance in the UC models;③Inhibiting the redundant production of NO in the colonic tissue;④Inhibiting the apoptosis of colonic epithdial cells;⑤Anti-inflammatory activity ,analgesic effect and antidiarrheal effects. |
