| Objective : Periodontitis is one chronic infective disease of periodontal supportive tissues, which caused by bacteria in dental plaque. And it is the main reason which leads to adult tooth loss.The hot spot of current studies is how to precipitate the cells which have regenerative potential to generate the functional periodontal tissues.The objective of this study is to observe the induction of osteoblastic differentiation in human periodontal ligament cells by Chinese medicine Gu Sui Bu, to research the cure feasibility of perodontitis by Chinese medicine Gu Sui Bu, and to lay a foundation of developing one new Chinese medicine of periodontitis by moderm technology.Material & Methods:Cell culture is established from human periodontal ligament by explant techniques.The 3th passage cells were identified by immunohistochemistry methods.To extract effective constituent of Chinese medicine Gu Sui Bu by distilled water. Cells were studied at 6th passage and were divided into 3 test groups at random. Each test group was added Chinese medicine Gu Sui Bu of different density(1×10mg/L,1×10~2mg/L,1×10~3mg/L) in DMEM medium. A group only cultured with DMEM was used as a blank control, and a group cultured with DMEM which included 200ug/L bone morphogenetic protein(BMP-2)was used as a positive control. The alkaline phosphatase(AKP) activity and osteocalcin(OC) concentration were measured respectively after 5 days.Result: Chinese medicine Gu Sui Bu of three density all significantly stimulated the AKP activity of human periodontal ligament cells, especially at the 1×10~2mg/L(P<0.01). Chinese medicine Gu Sui Bu had no effect at the OC concentration.Conclusions: Chinese medicine Gu Sui Bu could promote the osteogenic potential of periodontal ligament cells, and it has the worth of advance study. |
