| Object: To study the repair procedure of the rabbit nose cartilage defects with autogenous perichondrium /DBM complex in rhinoplasty and vocal fold wound healing.Methods: 1,sixteen adult New Zealand rabbits were randomly divided into two groups, created the both sides cartilage defects on the nose, The first group : autologous perichondrium/ allologous DBM complex was implanted into defect site in left side and autologous perichondrium were in right side, The second group: autologous perichondrium/ allologous DBM complex was implanted into the defect site in left side and autologous ear cartilage were in right side. All samples were collected at 10 weeks after transplantation. Gross observation histology and immunohistochemistry were used to evaluate the effect. 2,one-side recurrent laryngeal nerve was cut in ten adult New Zealand rabbits. Same lateral thyroid cartilage was creat windows, autologous perichondrium/ allologous DBM complex was implanted into laryngeal intrinsic tissue. All samples were collected at 6 weeks after transplantation. We used histology and immunohistochemistry to evaluate the configuration of DBM. 3,thirty adult New Zealand rabbits were randomly divided into six groups. The bilateral vocal cords wrere created injured with forceps. We used real-time PCR to test the expression of TGF-βmRNA.Results:1,the chondrocyte can be seen in the autologous perichondrium/ allologous DBM complex group. The morphology of cell is normal. Partly, the construction of DBM remained. There are few chondrocytes in the autologous perichondrium group. The cell and tissue of cartilage were normal in autologous ear cartilage group. Compared to the autologous ear cartilage group, the expression of s-100 protein no significantly decreased in autologous perichondrium/ allologous DBM complex group (P>0.05). the expression of TGF-β1 also significantly decreased (P<0.05). 2,There are no new chondrocyte in DBM implanted of 10- larynx, But the construction of DBE remain. 3,The expression of TGF-βmRNA has no change until 8 hours after of surgery of vocal fold,increased at 24hours and peaked at 72 hours. Conclusion: The autologous perichondrium / allologous Demineralized Bone Matrix(DBM) complex can induce the immigration of the chondrocyte of autologous perichondrium in the nose cartilage defect site. TGF-βmaybe play a significant role in the transplantation of cartilage and wound healing of vocal fold. |
