The Expression And Significance Of Integrin Beta1,VEGF And MMP-9 In Transitional Cell Carcinoma Of The Bladder

Background and Objects:As all the other tumors,the growth and progression of the bladder transition cell carcinoma have close correlation with the angiogenesis and invasion of the neoplasm,that process integrin family involve in.The abnormal expression of integrinα2,integrinα3 and integrinα6β4 had been found in many malignant tumors included BTCC,but the mechanism of action that integrins concerned about the growth and progression of BTCC is still ambiguity.We would like to find out how integrinβ1 play role in the angiogenesis,invasion and metastatic of BTCC,through detect and analysis the expression of with the clinicopathlogical index.Materials and Method:59 case of BTCC paraffin tissues sample collected from the patients underwent surgical resection,include 42 male and 17 female, average age is 62(34～80).According to the TNM stage classification of UICC and the grade classification of WHO,these samples could be classified into Tis 2 case,Ta 5 case,T1 18 case,T2 17 case,T3 15 case,T4 2 case;G1 16 case,G2 23 case,G3 20 case.In addition,we collected 15 case normal bladder tissue from the patients underwent the resection of benign prostatic hyperplasia as negative control.Using the SABC kit to detect the expression of integrinβ1,VEGF,MMP-9 in BTCC and normal bladder tissues,with PBS replaced the antibody as negative control.Buffy in cells is considered as positive cases,defined as positive were regarded by both staining intensity and the percentage of positive tumor cells.The enumeration data used Chi-squaer test apply for group comparison,and the measurement data used t test or variance analysis apply for group comparison.Results:1.The expression positive percentage of integrinβ1,VEGF,MMP-9 in BTCC was 45.76%,64.41%,55.93%,and MVD was 46.3±7.2.The expression positive percentage of integrinβ1,VEGF,MMP-9 in normal bladder tissues was 13.33%, 6.67%,6.67%,and MVD was 12.6±5.3.The difference between two group was significant.2.The difference of integrinβ1 expression positive percentage was significant between Tis-T1 stage and T2-T4 stage,G1,G2,and G3,simple and poly disease, diameter of tumor less or more than 3cm,age under or over 60y.The difference of VEGF expression positive percentage was significant between Tis-T1 stage and T2-T4 stage,G1,G2,and G3,simple and poly disease,diameter of tumor less or more than 3cm.The difference of MMP-9 expression positive percentage was significant between Tis-T1 stage and T2-T4 stage,G1,G2,and G3.The difference of MVD was significant between Tis-T1 stage and T2-T4 stage,G1, G2,and G3,simple and poly disease,diameter of tumor less or more than 3cm.3.There is a positive correlation between the expression of integrinβ1 and VEGF or between integrinβ1 and MMP-9(r=0.3703,r=0.3182).Conclusion:1.The expression of integrinβ1 increased in BTCC,and has correlation with the clinicopathlogical index such as stage or grade.2.Integrinβ1 might have cooperation with VEGF and MMP-9 in the angiogenesis and invasion of BTCC.
