Objective: Observe the expression change of early and late inflammation factors which are tumor necrosis factor-a(TNF-a)and high mobility group box chromosomal protein(HMGBl) in RAW264.7 macrophage after interfered with burning serum and Qing Huo Bai Du Yin serum(QHBDY serum) , find out QHBDY whether prevent the release of TNF-a and HMGB1Methods: Interfere the RAW264. 7 macrophage with normal SD rat serum , the mixture of normal SD rat serum and QHBDY feeding SD rat serum,the mixture of burning SD rat serum and normal SD rat serum ,the mixture of burning SD rat serum and QHBDY feeding SD rat serum ,use ELSA to observe the quantity of TNF-α, and western blot to find out the expression change of HMGB1 in nucleus ,cytoplastic and extracellular.Results : The quantity of TNF-αis very high in the mixture of burning SD rat serum and normal SD rat serum group, but it is lower in the mixture of burning SD rat serum and QHBDY feeding SD rat serum group.Two groups have statistical sense(p<0.05).It is little expression in normal SD rat serum group and the mixture of normal SD rat serum and QHBDY feeding SD rat serum group ,two groups have no statistical sense (p>0.05),but it has statistical sense comparing with two former groups. Western blot shows small quantity of HMGB1 expressed in normal SD rat serum groups and the mixture of normal SD rat serum and QHBDY feeding SD rat serum groups .Two groups have no statistical sense (p>0.05) .The quantity of HMGB1 increases in nucleus and cytoplastic in the mixture of burning SD rat serum and normal SD rat serum.and in can find extracellular 18h later. The quantity of HMGB1 also increases in the mixture of burning SD rat serum and QHBDY feeding SD rat serum groups.It is lower than burn groups. Two groups has statistical sense(p<0.05). The quantity of extracellular HMGB1 in the mixture of burning SD rat serum and QHBDY feeding SD rat serum groups is low.It depends time.The extracellular HMGB1can not be finded in normal SD rat serum groups and the mixture of normal SD rat serum and QHBDY feeding SD rat serum groups.Conclusion:Burning serum can stimulate RAW264.7 macrophage release early and late inflammation factors, TNF-αandHMGB1Herb serum of QHBDY can decrease the release of HMGB1 and TNF-αin RAW264.7 caused by burning serum.
