Objective: To reveal the activity and apoptotic condition of hematopoietic cells in the patients with psoriasis.Methods: Density gradient centrifugation and immunomagnetic beads method were used respectively to isolate marrow derived mononuclear cells and CD34+ cells.Withβ?actin as the internal control,the semi-quantitative detection of PKCβI expression of CD34+ cells from the patients with psoriasis and normal volunteers was measured by reverse transcription polymerase chain reaction(RT-PCR);Terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling(TUNEL method) was used to detect Apoptosis Index(AI) in the patients with psoriasis and normal control.Results: The relative quantity of KCβI mRNA expression of CD34+ cells was 1.12±0.16 in the patients with psoriasis,and that of normal controls was 0.90±0.14.PKCβI mRNA expression of D34+ cells in the patients with psoriasis was significantly higher than normal volunteers (t=4.24,P<0.05). Apoptosis Index of CD34+ cells in the patients with psoriasis was 5.9±1.0%. And Apoptosis Index was 3.4±0.7% in normal control. There was no significant difference between two groups(t=1.93, P>0.05).Conclusion: The biologic activity of hematopoietic cells were abnormal in patients with psoriasis.It suggests that hematopoietic cells play a role in the pathogenesis of psoriasis.
