Objective: To explore the therapeutic effects and possible mechanisms of action to ulcerative colitis by detecting the expression of nuclear factor-κB and thromboxane B2(TXB2) in rats with ulcerative colitis.Methods: An experimental rat model of ulcerative colitis was established by giving trinitro-benzene-sulfonic acid (TNBs), making salicylazosulfapyridine(SASP) as positive control. A total of 40 rats were randomly divided into four groups: the control group( group A), model group(group B), SASP treating group(group C), the traditional Chinese medicine treating group(group D). 14 days later, puting the 40 rats to death and getting blood plasma for detecting TXB2 using radioimmunoassay. Taking colon tissues for paraffin imbedding, and then getting some slice for HE dyeing and the other parts for detecting NF-κB using immunohistochemistry.Results: 1. The common conditions: The mouse of group B, C, D have diarrhea, less movement and their weight decresed after the model was established. The whole condition is the worst one week later. There was no obvious difference in growth of weight among group B, groupC and group D. 2. HE dyeing results: a great number of inflammatory cells infiltrated and crypt abscess emerged were observed under microscope in rats of B group. The inflammation was lessened after treating with SASP and traditional Chinese medicine. The ulcer repaired in the two groups. 3. Morphology and histology scores: the morphology and histology scores were significantly higher in group B than in group A. The scores of the two therapeutics groups were higher than group A, group B, respectively. There was no obvious difference in morphology and histology scores between group C and group D. 4. Expression of NF-κB and TXB2: the levels of NF -κB and TXB2 were significantly increased in group B. The two therapeutics groups showed higher expression of NF -κB and TXB2 than that in group B, respectively. There was no obvious difference in expression of NF -κB and TXB2 between group C and group D.Conclusion: 1.The levels of NF -κB and TXB2 were significantly increased in TNBs induced rat ulcerative colitis model. 2. The traditional Chinese medicine, Qingchangbaidu Decoction can down-regulation the expression of NF -κB and TXB2 in model rats. 3. The traditional Chinese medicine, Qingchangbaidu Decoction can decrease the area of impaired colon tissues of model rats. These studies indicate that Qingchangbaidu Decoction can resist ulcerative colitis by suppressing the expression of NF -κB and TXB2. |
