Obsjective:To investigate the change of brain histopathology,the expression of nuclear factor-κB(NFκB),interleukin-1(IL-1) and intercellular adhesion molecule-1 (ICAM-1) after focal cerebral ischemia-reperfusion in rats, and to discuss the probable mechanism of nerve growth factor(NGF)interaction effect.Methods:The model of focal cerebral ischemia-reperfusion in SD rats was established with suture-occluded method invented by ZeaLonga.54 SD rats were randomly divided into three groups(sham operated group,focal ischemia reperfusion model group, nerve growth factor group).Each group includes 18 SD rats.In All the groups,scores of neurological deficit,brain histopathology changes with HE staining in cortex ischemic region were observed,and immunohistochemistry was used to detect NFκB,IL-1,ICAM-1 in the same region at 24,48,72hours reperfusion after 2 hours ischemia.Results : (1)scores of neurological deficit:①sham operated group was no neurological deficit,②average scores of neurological deficit of focal cerebral ischemia reperfusion model group was 3, compared with sham operated group, it possess statistical significance (p<0.05);③average scores of group treated by NGF injection was 1, compared with model group, it possess statistical significance (p<0.05); (2)brain histopathology change in cortex ischemic region:①sham operated group was no brain histopathology change,②focal cerebral ischemia reperfusion model group:in the cores of ischemia,there were neuron damage:the amount of neuron decrease,cellular swelling,nuclear fragmentation,vascular was dilarationed around the core area with light microscope,③Compared with model group,we may find the same pathologic change in group treated by NGF ininjection,but the damage was less serious than focal cerebral ischemia reperfusion model group at the corresponding time;⑶the expression of NFκB,IL-1,ICAM-1 of the cortex ischemic region:①sham operated group was no change compared with correspond cortex,②the expression of NFκB of focal cerebral ischemia reperfusion model group were significantly higher than those in the sham operated group,its gray scale value after reperfusion 24h,48h,72h were 94.12±10.63,89.15±8.14,83.17±8.88 (all p<0.01) respectively,the expression peak value of IL-1,ICAM-1 of focal cerebral ischemia reperfusion model group were significantly higher than those in the sham operated group,their gray scale value after reperfusion 24h)were 170.62±5.24,126.36±4.77(all p<0.01)respectively,③the expression of NFκB of the NGF group treated were significantly lower than those in the focal cerebral ischemia reperfusion model group, its gray scale value at the corresponding time were 65.23±5.84,60.38±3.48,56.35±4.19(all p<0.05)respectively,the expression peak value of IL-1,ICAM-1 of the NGF group treated were significantly lower than those in the focal cerebral ischemia reperfusion model group,their gray scale value were 154.24±5.22,107.28±5.62(all p<0.05);Conclusion:⑴NGF may lessen SD rats neurological deficit symptom after focal cerebral ischemia-reperfusion;⑵NGF may lessen brain histopathology change in cortex ischemic region;⑶NGF may inhibit the expression of NFκB,IL-1 and ICAM-1,NGF may inhibit Inflammatory reaction,NGF was seem to have neuroprotective effect. |
