| [OBJECTIVE]By experiment of Methyl jasmonate on Hepatocellular carcinoma,we study the mechanism of Methyl jasmonate promote HepG-2 liver cancer cell proliferation, differentiation and apoptosis,further reveal the mechanism of liver cancer and to find a new,low toxicity,and highly effective drugs for Chemotherapy for Hepatocellular carcinoma.[METHODS]1.cultivation HepG2 cells in vitro,By MTT method to testing an effective concentration of methyl jasmonate;inverted microscope and HE staining to observe changes in shape of the cells;By Radioimmnnoassay method detect the AFP Change;Application of transmission electron microscope to observe the cell ultrastructure;flow cytometry detect the cell cycle and apoptosis.2.By RT-PCR detec thc Bcl-2,Bax mRNA expression;immunocytochemistry method detect the Bcl-2,Bax protein expression.[RESULTS]1.MeJA role of different times of the 50%inhibitory concentration(IC50)was 8.60 umol/L,optical microscope revealed that MeJA groups have some cells shedding and adherent cells decreased significantly,transmission electron microscope revealed that MeJA groups have some of cells connection and Microvilli disappear,and show that apoptotic bodies.MeJA group show the cell cycle G1 phase have significantly inhibit and some cell apoptosis,This shows that hypodiploid peak.2.Bcl-2 and Bax mRNA agarose gel electrophoresis results showed:MeJA group,ATRA group and MeJA + ATRA group of HepG2 cells have the Bcl-2 mRNA expression were significantly decreased(P<0.05)after 48h,but Bax mRNA expression levels were significantly higher(P<0.05);MeJA group,ATRA group and MeJA + ATRA group of HepG2 cells after 48h show Each group Bcl-2 protein in the cytoplasm of expression were significantly lower than the control group(P<0.01), but Bax protein levels were more significant increase in the control group(P<0.01).[CONCLUSIONS]Methyl jasmonate could inhibit HepG2 Cells Growth obviously,block cell cycle and promote apoptosis,inhibit the role of liver cancer;Methyl jasmonate through down Bcl-2 mRNA and protein levels,increased Bax mRNA and protein expression level, thereby induced apoptosis of HepG2 cells. |
